溶血对基于核磁共振技术代谢组学研究的影响

【摘要】 目的研究因溶血而造成的血浆标本中小分子代谢物组成及浓度的变化,探讨血浆标本溶血对代谢组学研究造成的影响。方法未发生溶血及发生溶血的健康男性血浆样本各10份,经核磁共振波谱仪检测后,应用SIMCA-P12.0软件对积分数据进行正交偏最小二乘判别分析及相关性分析。结果溶血组标本中涉及糖代谢、脂代谢及氨基酸代谢的多种小分子代谢物浓度发生改变。结论溶血可影响代谢组学分析结果,在实际操作中应该避免。

【Abstract】 Objective To investigate the effects of hemolysis on metabonomic results caused by the variation of plasma metabolite composition and concentration.Methods Nuclear Magnetic Resonance (NMR) based metabonomics was applied to analyze the plasma metabolic difference between plasma samples with hemolysis (n=10) and without hemolysis (n=10) obtained from healthy male subjects.OPLS-DA and Pearson′s correlative analysis was applied to the spectra data.Results The levels of metabolites involved in glucose,lipid and amino acid metabolism were changed in plasma samples with hemolysis.Conclusion Hemolysis could have effect on metabonomic results which should be avoided in studies.
