疏血通注射液对大鼠心肌缺血再灌注后心肌细胞凋亡和P53的影响

【摘要】 目的观察疏血通注射液(SXT)对大鼠心肌缺血再灌注后心肌细胞凋亡的影响。方法将18只大鼠随机分为假手术组、模型组和疏血通组,每组6只。通过结扎大鼠左冠状动脉前降支制成心肌缺血模型,疏血通组再灌注时即刻通过股静脉注射药物SXT(0.7mL/kg)24h。TUNEL免疫荧光染色观察心肌细胞凋亡,Westernblot检测P53表达。结果模型组心肌细胞凋亡显著,而疏血通组心肌细胞凋亡显著减少(P<0.01)。模型组P53表达显著高于假手术组、疏血通组(P<0.01)。结论 SXT可通过下调大鼠心肌缺血再灌注后心肌凋亡蛋白P53表达,从而减少心肌细胞凋亡。

【Abstract】 Objective To investigate the influence of Shuxuetong injection (SXT) on cardiomyocyte apoptosis in rats with myocardial ischemia/reperfusion. Methods Eighteen rats were randomly divided into sham group, model group and SXT group, with 6 rats of each group. The myocardial ischemia/reperfusion model was established by ligation of the anterior descending branch of the left coronary artery. SXT (0.7 mL/kg) was injected via femoral vein after reperfusion for 24 h in SXT group. Cardiomyocyte apoptosis and P53 expression were determined by TUNEL immunofluorescence and Western blot, respectively. Results Cardiomyocyte apoptosis decreased in SXT group compared with the model group (P<0.01). P53 expression in model group was superior to that in sham and SXT groups (P<0.01). Conclusion SXT can reduce the cardiomyocyte apoptosis via downregulation of P53 expression in rats with myocardial ischemia/reperfusion.
