两种钛表面成骨细胞附着静态与动态观察
Static and living observation on osteoblasts on two kinds of titanium surfaces
【摘要】 目的体外构建钛表面成骨细胞生长模型,对细胞生长情况进行动静态观察。方法将光滑钛片(光滑钛片组)及多孔海绵状钛(海绵钛组)处理为酸碱表面和钙磷沉积表面,接种成骨细胞,12 h、48 h实时观察,48 h后钛表面细胞固定,扫描电镜观察。结果光滑钛片组可见细胞充分延展附着,其中钙磷表面组细胞附着更多,可见细胞下钙磷沉积。海绵钛组可动态观察成骨细胞在海绵钛内立体生长,在钛珠间形成细胞桥。结论本研究构建形成钛表面成骨细胞生长体外模型,可进行钛表面细胞生长的实时观察与分析。
【Abstract】 Objective To establish titanium-osteoblast interface model in vitro,observe the osteoblast morphology in static and living.Methods The smooth and the sponge-like titanium were treated to acid-alkali surface and calcium & phosphorus surface respectively.Seeded osteoblasts on titanium surface and observed after 12 h and 48 h.Result On smooth titanium surface,osteoblasts extension adhesion was obsereved.There were more osteoblasts on calcium & phosphorus surface especially and calcium crystal could be seen beneath the cells.In sponge-like titanium group,osteoblasts cultured in 3-dimension and cell bridges could be seen in living.Conclusion Titanium-osteoblast interface model in vitro was successful established.It could be used to observe and analyse osteoblast on titanium in living and static.
