小干扰RNA沉默Dicer酶对人舌鳞癌细胞Tca-8113的增殖和细胞周期的影响
Influence of silencing Dicer by small interference RNA on the proliferation and cell cycle of Tca-8113 tongue squamous cell carcinoma cells
【摘要】 目的研究沉默Dicer酶的表达对人舌鳞癌细胞Tca-8113的增殖和细胞周期的影响。方法实验组转染靶向Dicer酶的siRNA序列;对照组转染无义序列siRNA;空白组细胞未做转染。采用RNA干扰技术沉默Tca-8113中Dicer酶的表达,通过荧光定量聚合酶链反应、蛋白质印迹法及免疫组织化学技术,检测Tca-8113转染小干扰RNA(small interference RNA,siRNA)后Dicer酶的mRNA和蛋白质水平的表达情况;应用噻唑蓝比色法及流式细胞术检测细胞增殖及细胞周期的变化。结果 Tca-8113细胞转染siRNA 24 h后,实验组Dicer酶mRNA表达量是空白组的0.39倍,表达水平下降,实验组与空白组或对照组比较,表达水平都有所下降,差异有统计学意义(P<0.05)。转染48 h后实验组蛋白质表达较空白组下调53.67%。Dicer酶表达下调后,Tca-8113细胞增殖加速,G1期细胞百分率减少,S期和G2期细胞百分率增多。结论沉默Dicer酶的表达可加快细胞周期从G1期向S期和G2期进展,促进Tca-8113细胞的生长。
【Abstract】 Objective To study the influence of silencing Dicer by small interference RNA(siRNA) on the proliferation and cell cycle of Tca-8113 tongue squamous cell carcinoma cells.Methods The silencing effect of RNA interference on Dicer expression was evaluated by quantitative real time PCR,Western blot and immunohistochemistry staining.The cell proliferation rate and cell cycle kinetics were detected by MTT assay and flowcytometry,respectively.Results The siRNA targeting Dicer suppressed the expression of Dicer in Tca-8113 cells.Meanwhile,the proliferation activity was significantly enhanced in cells transfected with Dicer siRNA compared to the controlled cells.Conclusion The preliminary findings suggest that lower expression of Dicer may play an oncogenic role.
