气相色谱检测发酵食品中MVOCs的条件优化
Conditions optimization of detection on MVOCs in fermentation food by Gas Chromatography
【摘要】 以近10种发酵食品中常见且较难分开的有机物质作为检测对象,优化气相色谱检测时的载气柱流量和柱温2个影响分析时间和分离效果的重要条件,并在谱峰归属试验和重复性试验中讨论保留时间、相对保留时间、峰面积和相对峰面积在定性定量分析中的作用。结果表明:载气流量为2.5 mL/min,柱温条件为50℃(保持3min)→100℃(3℃/min)→200℃(15℃/min,保持10 min)的色谱条件较为适宜。在此条件下,混合物中各组分可以完全分离且分析时间为36.3 min,较为适中。通过谱峰定性试验和重复性试验分析得出定性定量方法,用乙醚溶剂作内标物,采用相对保留时间定性和相对峰面积定量较好。所选几种物质中低沸点物质检出限较低,高沸点物质检出限较高,因此需要对样品进行浓缩处理才能满足检测要求。
【Abstract】 Nearly 10 kinds of organic matters which were common in fermentation food and not easy to be separated from each other in GC were chosen as test objects. Carrier gas flow rate and column temperature were two main conditions of gas chromatographic detection, which were optimized in this experiment. And the role of the retention time, relative retention time, peak area and relative peak area in the qualitative and quantitative analysis was also discussed. Results showed that the carrier gas flow rate was 2.5 mL/min, the oven temperature conditions of 50℃(keep 3 min) to 100℃(3℃/min) to 200℃(15℃ /min, keep 10 min) were suitable chromatographic conditions. And under the conditions, the components in the mixture could be completely separated and the analysis time was 36.3 min, which was relatively moderate. Using diethyl ether as internal standard, and adopting relative retention time and relative peak area for quantitative and qualitative analysis was better. For selected several kinds of substances, low detection limit could be gotten for low boiling point, and high boiling point substances’ detection limit was high. This result indicated that the samples needed to be concentrated to meet the testing requirements.
