TCDD宫内暴露对子鼠肝细胞和亚细胞结构的毒性作用
Effects of intrauterine exposure to TCDD on hepatocytes and subcellular structure in livers of neonatal rats
【摘要】 目的探讨动物妊娠早期接触2,3,7,8-四氯二苯并二噁英(TCDD)后对出生3 d的子鼠肝的毒性作用。方法孕13 d SD大鼠随机分为空白对照组(n=6)、玉米油组(n=6)和染毒组(n=18),即在妊娠13 d分别给予玉米油和用玉米油配制的5、100、500 ng·kg-1·d-11.0 mL不同剂量的亚组(n=6)至分娩,取每组出生3 d的雌性小鼠的肝(n≥6)。石蜡切片,HE染色,光镜观察,电镜观察。结果空白对照组和玉米油组的细胞结构和亚细胞结构均未见病理性改变。TCDD宫内暴露染毒组中,5 ng组肝索结构排列紊乱,胞浆水肿明显;而100 ng组的肝细胞体积明显变小,胞浆内有大量脂肪滴溶解后的空泡,并且有少量细胞核固缩;500 ng组肝细胞肿胀、脂肪变更为明显,胞浆浊样加剧,胞浆空泡化。对亚细胞结构的毒害作用主要是使线粒体线的脊排列紊乱、溶解。结论TCDD宫内暴露对子鼠的肝造成了病理性的改变,并且随着染毒剂量的增加,病理改变越明显。
【Abstract】 Objective To study the toxic effect of liver structure in neonatal rat after intrauterine exposure to 2,3,7,8- tetrachlorodibenzo-p-dioxin(TCDD) in early pregnancy.Methods Pregnant 13 d SD rats were randomly divided into control group(n=6),corn oil group(n=6) and poisoned group(n=18) which was given corn oil and TCDD(dissolved by corn oil in 5 ng,100 ng,500 ng·kg-1·d-11.0 mL) through dose gavages respectively during pregnant from 13d to delivery.Then getting livers(n ≥6) from female neonatal rats which were born 3 days in paraffin section.HE stain,and light microscopy observation,electron microscope observation.Results There weren’t any pathological features founded in rats between control group and corn oil group.In poisoned group,the hepatic cord was disordered arrangement and the cytoplasm swelling significantly in the dosage level of 5ng group.In 100 ng group,there were significantly decreasing in the volume of liver cell.A large number of vacuoles coming from dissolved fat droplets in cytoplasm and a small amount of karyopyknosis can be observed.In 500 ng group,swelling cells can be observed,which had fatty change,cloud-like change and obvious vacuoles in the cytoplasm.The toxicity of the subcellular of liver mainly happened in the ridge of the mitochondrial,which become disordered and dissolved.Conclusion Intrauterine exposure to TCDD caused pathological changes of the liver in neonatal rats and the damage is more obvious as the toxin dosage was increased.
