荔枝核总黄酮对肝纤维化大鼠肝组织Toll样受体2、4表达的影响

【摘要】 目的探讨荔枝核总黄酮(TFL)对胆管阻塞型肝纤维化大鼠肝组织Toll样受体(TLR)2、TLR4表达的影响。方法雄性SD大鼠80只,随机分为正常对照组(A组)、模型组(B组)、TFL小剂量组(C组)、TFL大剂量组(D组)。除A组外,其余各组均采用胆总管结扎法制备肝纤维化大鼠模型。在造模后第2天开始,A、B组给予生理盐水灌胃,C、D组分别给予100、200 mg/(kg·d)的TFL灌胃,均1次/d,共4周,第4周末处死全部大鼠,留取肝脏组织。HE、Masson染色观察大鼠肝组织病理学改变,免疫组化法及免疫印迹法检测TLR2、TLR4在4组大鼠肝组织内的表达。结果与B组比较,D组肝组织肝纤维化程度明显减轻,胶原表达明显减少(P<0.01);D组大鼠肝组织中TLR2和TLR4蛋白明显减少(P<0.01),C组介于B组和D组之间,但与B组比较差异无统计学意义(P>0.05)。结论 TLR2和TLR4蛋白表达增加可能与胆总管结扎大鼠肝纤维化的发生发展有关,200 mg/(kg·d)TFL能抑制肝纤维化大鼠TLR2、TLR4的表达,并改善大鼠肝组织纤维化程度。

【Abstract】 Objective To investigate the effects of total flavone of litchi chinensissonn( TFL) on the expression of toll- like receptor- 2( TLR- 2) and TLR- 4 in rats with hepatic fibrosis. Methods Eighty males SD rats were randomly divided into four groups: sham- operated group( Group A),bile duct ligation group( Group B),low- dose TFL group( Group C),and high- dose TFL group( Group D). The liver fibrosis rat model was induced by common bile duct ligation( CBDL). TFL lavage was given to subjects in Group C and D,while replacing saline was givento those in Group A and B from the 2nd day of operation for 4 weeks. Histopathological changes in the livers were studied by hematoxylin- eosin( HE) staining and massontrichrome( MT) examination. The expression of TLR- 2 and TLR- 4 in liver tissues was assessed. Results The collagen intensity score and area in Group D was significantly lower than those in Group B( P <0. 01),with significantly lower expressions of TLR- 2 and TLR- 4( P < 0. 01). Conclusion The up- regulation of TLR- 2 and TLR- 4 is correlated with the pathogenesis of CBDL- induced liver fibrosisrats,which could alleviated by high dose TFL.
