RNA干扰技术沉默IGF-1R效应对子宫内膜癌细胞增生及裸鼠成瘤的抑制作用
The inhibited effect of RNA interference technology silencing IGF-1R on endometrial carcinoma proliferation and tumorigenesis in nude mice
【摘要】 目的研究靶向胰岛素样生长因子受体-1(IGF-1R)的小干扰RNA(siRNA)对子宫内膜癌生长的抑制作用。方法采用实时定量PCR(real-time PCR)和蛋白印迹法(Western-blot)测定RNA干扰后IGF-1R的表达水平;采用四甲基偶氮唑蓝比色法(MTT)检测细胞增殖能力;流式细胞仪检测细胞凋亡及周期改变;裸鼠成瘤实验测定IGF-1R基因表达下调后子宫内膜癌细胞在体内致瘤能力的改变。结果靶向IGF-1R基因的siRNA可以有效抑制IGF-1R基因的表达,使IGF-1R mRNA表达减少85.0%,蛋白表达减少91.4%;流式细胞术检测到转染后细胞凋亡率明显升高,发生G2/M期阻滞;裸鼠体内成瘤能力降低。结论 siRNA介导的IGF-1R基因下调能明显抑制子宫内膜癌的体内外生长。
【Abstract】 Objective In order to explore the inhibited effect of RNA interference targeting insulin-like growth receptor-1(IGF-1R) on endometrial carcinoma growth and tumorigenesis in nude mice, we constructed siRNA targeting IGF-1R and transfected HEC-1B cells. Methods We constructed siRNA targeting IGF-1R and transfected HEC-1B cells. Real-time PCR and western-blot were used to detect the expression of IGF-1R after transfection.Cell proliferation ability was determined by MTT method, change of cell apoptosis and cell cycle was detected by flow cytometryt.Nude mice model was used to measure the tumorigenesis ability of endometrial carcinoma after down-regulation of IGF-1R expression.Results RNA interference targeting insulin-like growth receptor-1 could effectively inhibit IGF-1R expression, which made mRNA level reduced by 85.0%, protein level reduced by 91.4%, cell apoptosis after transfection was significantly increased measured by flow cytometry, G2/M phrase was blocked, and the tumorigenesis ability of nude mice inoculated with endometrial carcinoma HEC-1B cells was decreased. Conclusion Down-regulation of IGF-1R mediated by siRNA could effectively inhibit endometrial carcinoma growth both in vivo and in vitro.
