吲哚胺2,3-二氧化酶对小鼠同种异体肝细胞脾内移植的作用

【摘要】 目的研究携带吲哚胺2,3-二氧化酶(IDO)的小鼠肝细胞在同种异体脾内的表达及对脾淋巴细胞的凋亡作用。方法分离BABL/c小鼠肝细胞,肝细胞脾内移植(BABL/c→C57BL/6),分为4组:Ⅰ组为携带IDO组;Ⅱ组为1-甲基色氨酸组;Ⅲ组为空壳腺病毒组;Ⅳ组为单纯肝细胞移植组。移植术后第2,5d取脾脏免疫组织化学检测IDO表达情况,TUNEL检测脾脏淋巴细胞凋亡情况。结果携带IDO腺病毒转染肝细胞后,能有效表达IDO。免疫组织化学显示,Ⅰ、Ⅱ组脾脏有IDO阳性的肝细胞。与其他组比较,移植术后第2,5dⅠ组淋巴细胞凋亡差别有统计学意义(P<0.05),Ⅱ、Ⅲ、Ⅳ组之间比较,差别无统计学意义(P>0.05)。结论携带IDO的肝细胞能在小鼠同种异体脾内稳定表达,并能诱导脾内淋巴细胞的凋亡。

【Abstract】 Objective To evaluate the effects of Indoleamine 2,3-dioxygenase(IDO)on the mice allogeneic hepatocytes transplanted into spleen. Methods BABL/c mice hepatocytes were isolated and transplanted into C57BL/6 mouse and divided into IDO group(I group),IDO+1-methyl tryptophan(1-MT)group(Ⅱ group),shell adenovirus group(Ⅲgroup)and hepatocyte group(Ⅳgroup). The IDO was detected by immunohistochemistry and lymphocyte apoptosis of spleen were detected by TUNEL on the day 2,5after transplantation. Results Hepatocytes effectively expressed IDO after transfection.Immunohistochemistry showed that IDO-positive hepatocytes were observed in Ⅰ and Ⅱ group. Compared with other groups,the difference of lymphocyte apoptosis inⅠgroup was statistically significant on the day 2,5after transplantation(P<0.05),no difference were observed between Ⅱ,Ⅲ and Ⅳ groups(P>0.05). Conclusion Mice allogeneic hepatocytes transfected with IDO can stably express IDO and induce lymphocyte apoptosis in spleen.
