藏红花素对CNE2细胞的增殖及迁移抑制作用
The Proliferation and Migration Inhibition and Mechanism of Crocin on CNE2 Cells
【摘要】 目的探讨藏红花素(crocin)对人鼻咽癌细胞株CNE2细胞的增殖和迁移抑制作用及其可能的作用机制。方法使用高倍显微镜观察不同浓度crocin作用后的细胞形态变化,CCK-8法测定细胞增殖能力的改变,Transwell小室实验分析细胞迁移能力变化,RT-PCR技术测定bi-1基因mRNA表达变化。结果藏红花素作用于CNE2细胞24h后,可抑制其增殖和迁移能力,且呈剂量依赖关系,但当药物浓度超过0.8 mmol/L时,细胞形态发生明显改变,细胞死亡增加。RT-PCR结果显示,不同浓度的藏红花素作用于CNE2细胞24h后,bi-1基因的mRNA表达明显下调。结论藏红花素对CNE2细胞有增殖和迁移抑制作用,并可影响抗凋亡基因bi-1的表达,其机制可能是通过调控bi-1基因的表达实现的。
【Abstract】 Objective To investigate the effect of crocin on the proliferation and migration of CNE2 cells,and explore the underlying molecular mechanism.Methods Microscope was used to observe morphological changes of CNE2 cells after cultured with different concentrations of crocin.CCK-8 experiment was used to examine the inhibition of crocin on the proliferation of CNE2 cells.Transwell was used to determine the migration ability of CNE2 cells.RT-PCR reaction was used to test mRNA expression of bi-1 gene.Results After using different concentrations crocin for 24 hours,CNE2 cells was suppressed both in proliferation and migration ability in a dose-dependent way.But when the crocin concentration is higher than 0.8 mmol/L,cell morphology would significantly change,or even died.RT-PCR reaction showed that bi-1 gene mRNA expression in CNE2 cells was down-regulated after being treated by different concentrations of crocin.Conclusion Crocin can inhibit the proliferation and migration of CNE2 cells,and can affect the expression of anti-apoptosis gene bi-1.Further experiments illustrated that the inhibition is probably induced by regulation of bi-1 gene.
