盐酸纳美芬对大鼠颅脑创伤后Survivin表达的影响

【摘要】 目的观察盐酸纳美芬对大鼠颅脑创伤后生存素(Survivin)表达的影响,探讨其脑保护机制。方法 78只Wistar大鼠随机分为假手术组(n=6)、颅脑创伤组(n=36)及纳美芬治疗组(n=36)。采用自由落体法建立大鼠颅脑创伤模型,应用Epics XL流式细胞仪检测细胞凋亡情况并应用免疫组织化学法检测Survivin和半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)蛋白表达变化。结果与假手术组相比,颅脑创伤组细胞凋亡率及Survivin、Caspase-3蛋白表达水平明显升高。盐酸纳美芬可促进Survivin表达,并通过相关性分析,其表达水平和Caspase-3表达具有负相关性。结论盐酸纳美芬可能通过增加抗凋亡蛋白Survivin表达来抑制Caspase-3激活,减少颅脑创伤后细胞凋亡,发挥神经保护作用。

【Abstract】 Objective To investigate the effect of Nalmefene on survivin expression in rats after traumatic brain injury(TBI),and explore the mechanisms of neuro-protection by Nalmefene.Methods Seventy-eight adult Wistar rats were randomly divided into sham-operation group(n = 6),traumatic brain injury(TBI) group(n = 36) and Nalmefene group(n = 36).The experimental TBI model was established by Feeney’s method.Samples were obtained after injury for measuring the cells apoptosis by Epics XL Flow Cytometer and inspecting the expression of survivin and caspase-3 proteins by an immunochemical method.Results Compared with the sham-operation group,the apoptosis of cells and expression of survivin and caspase-3 proteins were significantly increased in the injured brain after TBI.Nalmefene significantly increased the expression of survivin,which was negatively correlated with the activity of caspase-3 according to the correlation analysis.Conclusion Increasing the expression of survivin by nalmefene might be one of the mechanisms in regulating caspase-3 level and inhibiting neuronal apoptosis in cortex and exerting protective function to neurons.
