酶催化分解双氧水的动力学性能研究
Research on the Dynamic Performance of Enzyme Catalytic Decomposition of Hydrogen Peroxide
【摘要】 在经典实验的基础上,以猪肝中提取的酶作为催化剂,与传统的催化剂相比,酶受外在因素影响更为敏感。实验过程中选取3个因素(温度、浸取时间、pH)作为变量,进行初步定量分析,并以此设计正交实验,为动力学参数的测定提供良好的依据。在动力学参数的处理上,采用的是Guggenheim法处理过氧化氢催化分解反应动力学数据,此法不用测量V∞,可减小误差。当浸取时间为8小时,温度为37℃,pH为6.8时,酶催化过氧化氢分解效率达到最高;在3个考察因素中,pH对酶催化效率影响最大。酶催化双氧水分解的动力学数据表明,其反应速率常数变化不规律,受环境影响较大。
【Abstract】 Based on the classic experiment,enzyme extracted from pork liver can be more easily affected by external factors as catalyst than the traditional enzyme Three factors(temperature,le-aching time,pH)were selected in the experiment as variables.The preliminary quantitative analysis of the three factors and the orthogonal design experiment have been studied,which provides a good basis for the determination of kinetic parameters.Using the Guggenheim method to treat catalytic decomposition of hydrogen peroxide reaction kinetics data,this method does not need to measure the value of V∞to reduce errors.If the extraction time is 8 hours the temperature is 37 ℃,and the pH is 6.8,enzyme catalytic decomposition of hydrogen peroxide can achieve maximum efficiency.However,among the three factors,the effect of pH value influences the catalytic efficiency of the enzyme most.The dynamics data of the enzyme catalysis hydrogen peroxide solution decomposition indicates that its process is influenced bythe environmental factors.
