慢性心衰大鼠下丘脑室旁核谷氨酸脱羧酶对交感神经活动的影响
Downregulation of Glutamic Acid Decarboxylase 67 in Paraventricular Nucleus Contributes to Sympathoexcitation in Rats with Chronic Heart Failure
【摘要】 本文证实慢性心衰大鼠下丘脑室旁核(PVN)谷氨酸脱羧酶对交感神经活动的影响。冠脉结扎法诱导大鼠慢性心衰,在PVN微量注射谷氨酸脱羧酶抑制剂烯丙基甘氨酸,观察动脉血压、心率和肾交感神经活动的变化,结果显示假手术组和心衰组大鼠心率、血压和肾交感神经放电均有升高(P<0.05),但心衰组升高幅度与假手术组相比显著减小(P<0.05)。Western blot实验法观察PVN内谷氨酸脱羧酶蛋白表达量变化,结果显示与假手术组比,心衰组大鼠PVN内谷氨酸脱羧酶67(GAD67)表达下调(P<0.05),但谷氨酸脱羧酶65(GAD65)表达无明显变化(P>0.05)。结果提示心衰大鼠PVN的谷氨酸脱羧酶变化对交感神经的作用发生钝化,其机制可能与PVN内谷氨酸脱羧酶67表达量下调有关。
【Abstract】 The goal of this study was to determine the role of glutamate decarboxylase(GAD) in modulating the tonic inhibitory influence on the paraventricular nucleus(PVN) in rats with heart failure(HF).HF animal model was induced by coronary artery ligation.Unilateral microinjection of GAD inhibitor allylglycine into the PVN produced a dose-dependent increase in renal sympathetic nerve activity(RSNA),heart rate(HR)and blood pressure(BP)in both sham and HF rats.There was a blunted increase in these parameters in HF rats compared with the sham rats.The results of Western blot show that GAD67 protein expression levels in the PVN were significantly decreased in HF rats compared with sham rats,but there were no significantly different GAD65 protein expression levels.Our results indicate that GAD plays the tonic inhibitory influence on PVN is blunted and the mechanism may be involved in the downregulation of GAD67 in PVN.
