滋补脾阴方药对脾阴虚糖尿病大鼠大脑皮质胰岛素信号通路的影响

【摘要】 目的:通过观察大鼠大脑皮质胰岛素信号通路的变化,探讨滋补脾阴方药(ZBPYR)的作用机制,为糖尿病认知功能障碍的治疗提供新思路和新线索。方法:将大鼠随机分为空白对照(cont)组,糖尿病(DM)组,脾阴虚(pi)组,脾阴虚糖尿病(piDM)组,滋补脾阴方药治疗(ZBPYR)组5组。采用Western blotting方法观察胰岛素受体底物1(IRS-1)及蛋白激酶B(PKB/Akt)蛋白表达变化,以判定是否出现胰岛素信号转导障碍。结果:DM组、pi组、piDM组p-IRS-1(丝氨酸磷酸化)表达较cont组增加(P<0.05),DM组、piDM组大鼠p-Akt表达减弱(P<0.05),ZBPYR组p-IRS-1(丝氨酸磷酸化)较DM组、piDM组减弱(P<0.05),p-Akt表达较DM组增加(P<0.05)。结论:DM组、pi组、piDM组大鼠大脑皮质中发生了胰岛素抵抗,ZBPYR具有纠正脑胰岛素抵抗的作用。

【Abstract】 Objective:By observation of the changes of insulin signaling pathway in cortex of rats,to explore the mechanism of Zibu Piyin Recipe(ZBPYR),providing new ideas and new clues for treatment of diabetes-associated cognitive decline(DACD).Methods:The rats were randomly divided into five groups:cont group,DM group,pi group,piDM group and ZBPYR group.The protein level of insulin receptor substrate 1(IRS-1) and protein kinase B(PKB/Akt) which involved in insulin signaling were observed by Western blotting in cortex to determine whether insulin signaling obstacles in cortex of rats.Results: The expression of p-IRS-1(serine phosphorylation) of DM group,pi group and piDM group was increased compared with cont group(P<0.05),the expression of p-Akt of DM group and piDM group was decreased(P<0.05),the expression of p-IRS-1(serine phosphorylation) of ZBPYR group was decreased compared with DM group and piDM group(P<0.05),while the level of p-Akt increased(P<0.05).Conclusion:Insulin resistance occurs in cortex of rats of DM group,pi group and piDM group,ZBPYR could improve insulin resistance in brains.
