健脾益肾活血法对ITP小鼠淋巴细胞Fas/FasL蛋白表达的影响

【摘要】 目的:观察健脾益肾活血法对特发性血小板减少性紫癜(ITP)模型小鼠淋巴细胞Fas/FsaL蛋白表达的影响。方法:BALB/c小鼠随机分为空白对照组、模型对照组、PA对照组和JYH低、中、高剂量组,各组连续灌胃14d。于实验第5天开始,除空白对照组外,各组连续腹腔注射豚鼠抗小鼠血小板血清(GP-APS)复制ITP模型。检测外周血血小板(PLT)、平均血小板体积(MPV)、血小板分布宽度(PDW)、血小板压积(PCT),淋巴细胞Fas/FsaL表达阳性率。结果:与空白对照组比较,模型对照组小鼠PLT数量明显减少(P<0.05),与模型对照组比较,PA对照组和JYH低、中剂量组PLT数量明显增加(P<0.05);模型对照组MPV均值明显高于空白对照组(P<0.05),与模型对照组比较,JYH中剂量组MPV明显降低(P<0.05);模型组小鼠淋巴细胞Fas+/FsaL+表达与空白对照组比较显著上调(P<0.05),与模型对照组比较,JYH低、中、高剂量组及PA对照组可不同程度下调其Fas+/FsaL+表达(P<0.05)。结论:健脾益肾活血法对ITP小鼠淋巴细胞Fas/FsaL异常表达具有调节作用。

【Abstract】 Objective: To observe the effect of Jiangpi Yiqi and Huoxue treatment principle on lymphocytes Fas and FsaL protein expression of ITP model mice.Methods: The BALB/c mice were randomly divided into 6 groups(Normal control group,Model control group,PA control group,JYH low,middle and high dose groups).Mice were continuously given irrigated gastrom for 14 days.On the fifth day of test,mice were injected with Guinea pigs anti mice platelet serum(GP-APS) solution into the celiac of mice for duplicating the ITP model except normal control group.Detect peripheral blood PLT,PCT,MPV,PDW and Lymphocyte Fas/FsaL express positive rate.Results: The PLT was significantly decreased in model control group than that in normal control group(P<0.05).The PLT of mice in treatment groups were higher than that in model group,among the rest the PA control group and JYH low,middle dose group had significant difference(P<0.05);Model control group was higher than the mean MPV normal control group(P<0.05),compared with model control group,MPV of every other group was reduced in different degrees,one of JYH dose group has significant difference;Lymphocytes Fas+/FsaL+ expression of each model group were significantly increased compared with normal control group(P<0.05).Compared with model control group,JYH treatment group could down regulate the Fas+/FsaL+ expression in different degrees(P<0.05).Conclusion: Jiangpi Yishen and Huoxue treatment principle had regulatory role on lymphocytes Fas/FsaL abnormal expression of ITP model mice.
