‘雪梅’未成熟合子胚体胚发生与植株再生
Somatic embryogenesis and plant regeneration from immature zygotic embryos of Prunus mume ‘Xue Mei’
【摘要】 以‘雪梅’未成熟合子胚为外植体,探讨了不同采样时期和不同浓度2,4-D与6-BA组合对体胚发生的影响。结果表明:盛花后约59 d的‘雪梅’未成熟合子胚,暗培养60 d后,在12MS+2,4-D 1.0 mgL+6-BA 0.5 mgL上培养时体胚诱导率达最高(58.3%),在12MS+2,4-D 1.0 mgL+6-BA 0.2 mgL上培养时体胚诱导数最高(达3.6个);体胚在增殖培养基12MS+6-BA 0.1 mgL+NAA 0.05 mgL中产生次生体胚,在12MS+6-BA 1.0 mgL+NAA 0.1 mgL+GA30.5 mgL培养中萌发,再生出植株。
【Abstract】 Using immature zygotic embryos of ‘Xue Mei’as explants,the effects of different sampling dates and combinations of 2,4-D and 6-BA on somatic embryogenesis were investigated. The results indicated that the highest induction rate of 58.3% was obtained from immature embryos of‘Xue Mei’collected on the 59th day after flowering and incubated for 60 d on the 1/2 MS + 1.0 mg/L 2,4-D + 0.5 mg/L 6-BA in the dark. The maximum average number of somatic embryos could reach 3.6 on the 1/2 MS + 1.0 mg/L2,4-D + 0.2 mg/L 6-BA. Secondary somatic embryos could be produced when primary somatic embryos were cultured on proliferation medium 1/2 MS + 0.1 mg/L 6-BA + 0.05 mg/L NAA,and plantlets generated from somatic embryos on germination medium 1/2 MS + 1.0 mg/L 6-BA + 0.1 mg/L NAA + 0.5mg/L GA3.
