两种可注射磷酸钙骨水泥的细胞相容性研究
Cell biocompatibility of two injectable calcium phosphate cements
【摘要】 目的检测可注射α-磷酸三钙/磷酸钙骨水泥和β-磷酸三钙/磷酸钙骨水泥材料的体外细胞相容性。方法将小鼠成骨前体细胞株MC3T3-E1分别接种于α-磷酸三钙/磷酸钙骨水泥和β-磷酸三钙/磷酸钙骨水泥的固化块上,应用扫描电镜观察细胞在材料上的粘附情况。通过四唑盐比色实验(MTT法)和碱性磷酸酶(alkaline phosphatase,ALP)活性测定,检测细胞的增殖、分化能力。结果 MC3T3-E1在α-磷酸三钙/磷酸钙骨水泥和β-磷酸三钙/磷酸钙骨水泥固化块上能较好地粘附,在β-磷酸三钙/磷酸钙骨水泥固化块上MTT吸光值随时间延长增长幅度较大,且ALP活性高于α-磷酸三钙/磷酸钙骨水泥。结论 MC3T3-E1的增殖和分化能力在β-磷酸三钙/磷酸钙骨水泥上高于α-磷酸三钙/磷酸钙骨水泥。
【Abstract】 Objective To examine the cell biocompatibility of injectable α-TCP/CPC and β-TCP/CPC in vitro.Methods The mouse preosteoblastic cells(MC3T3-E1) were cultured on setting discs of β-TCP/CPC and α-TCP /CPC.The adhesion,proliferation and differentiation of the MC3T3-E1 were observed by SEM,MTT assay and the activity of alkaline phosphatase(ALP).Results The MC3T3-E1 could adhere on the discs of α-TCP/CPC and β-TCP/CPC,the MTT value of β-TCP/CPC increased rapidly with time and the activity of ALP of β-TCP/CPC was higher than that of α-TCP/CPC.Conclusion The proliferation and differentiation of the MC3T3-E1 on β-TCP/CPC discs were higher than those of α-TCP/CPC.
