全反式维甲酸对兔颈动脉内膜损伤病灶中内膜增生和IL-8及PECAM-1表达影响

【摘要】 目的观察全反式维甲酸(ATRA)对兔颈动脉内膜损伤病灶中内膜增生和白细胞介素-8(IL-8)、血小板内皮细胞黏附分-1(PECAM-1)表达的影响。方法将54只新西兰大白兔随机分为假手术组、对照组和治疗组3组。对照组和治疗组给予高脂饮食,2周后,用空气干燥法制作颈动脉内膜损伤病灶模型,治疗组于术前3d开始给予ATRA灌胃。假手术组给予普通饮食,2周后分离暴露颈动脉但不损伤内膜。于术后第7、14、28天分别处死各组动物,取损伤血管进行形态学观察,采用免疫组化法检测IL-8和PECAM-1的表达水平。结果假手术组未见内膜增生,IL-8无表达,PECAM-1仅有微量表达;对照组术后7d内膜开始增生,14、28d时内膜增生明显,管壁有粥样斑块形成,管腔狭窄,IL-8和PECAM-1表达量增多;治疗组内膜增生相对于对照组较轻,14和28d时内膜面积显著低于对照组(t=4.041、3.896,P<0.05),治疗各组IL-8和PECAM-1的阳性表达指数也显著低于对照组(t=3.368~4.421,P<0.05)。结论 ATRA可通过抑制IL-8和PECAM-1的表达从而抑制动脉内膜损伤病灶中的内膜增生,维持粥样斑块的稳定性。

【Abstract】 Objective To observe the influence of all-trans retinoic acid(ATRA) on the intimal hyperplasia and expressions of IL-8 and PECAM-1 in injured focus of carotid artery of rabbits. Methods Fifty-four New Zealand rabbits were equally randomized to sham-operation,control and treatment groups.The rabbits in the control and treatment groups were fed with high fat diet,two weeks later,a model of carotid-intimal-injury was created by air desiccation,those in the treatment group received lavage of ATRA daily,three days before operation.The rabbits in the sham-operation group were fed with general diet for two weeks,and then the carotid arteries were dissected and exposed,but the intima was not injured.The animals were sacrificed on days 7,14 and 28,respectively,after surgery.The injured blood vessels were taken for morphologic observation,the expressions of IL-8 and PECAM-1were detected by immunohistochemical method. Results In the sham-operation group,neither intimal hyperplasia nor expression of IL-8 was observed,and the expression of PECAM-1 was minimal.In the control group,neointimal began on day 7,which became obvious on days 14 and 28,with formation of atherosclerotic plaque and stricture of lumen,and the expressions of IL-8 and PECAM-1 increased.In the treatment group,neointimal was less and its area on days 14 and 28 was smaller than that in the control(t=4.041,3.896;P<0.05),and the expressions of IL-8 and PECAM-1 in each treatment group was also lower(t=3.368-4.421,P<0.05). Conclusion ATRA can inhibit the intimal hyperplasia of injured tunica intima of artery through inhibiting the expressions of IL-8 and PECAM-1,and keeping the atheromatous plaque stable.
