中华鳖和杂交鳢血清免疫球蛋白免疫印迹和酶联免疫分析

【摘要】 [目的]探讨中华鳖和杂交鳢血清免疫球蛋白的同源性及其含量。[方法]采集中华鳖和杂交鳢血清与25种商品二抗共同抚育,进行免疫印迹及酶联免疫分析。[结果]中华鳖和杂交鳢血清可与兔抗鸡IgM、兔抗猪IgG、兔抗鸡IgG、兔抗大鼠IgM、兔抗猴IgG和羊抗人IgG等6种二抗发生免疫印迹反应。利用鸡IgG、鸡IgM、猪IgG、大鼠IgM、猴IgG和人IgG的ELISA试剂盒测定了中华鳖和杂交鳢血清免疫球蛋白水平,中华鳖血清免疫球蛋白浓度为28.5~215.4μg/L,杂交鳢血清免疫球蛋白浓度为24.5~201.5μg/L。[结论]该研究能够丰富中华鳖、杂交鳢特异性免疫系统的研究,对中华鳖、杂交鳢的健康养殖有一定的指导意义。

【Abstract】 [Objective] The research aimed to study the homology and content of serum immunoglobulin in Trionyx sinensis and Ophiocephalus argus Cantor(♂)×Ophiocephalus maculatus(♀).[Method] The serum of T.sinensis and O.argus Cantor(♂)×O.maculatus(♀) was sampled to cultivate with 25 commodities secondary antibodies for Western blot and ELISA analysis.[Result] The serum of T.sinensis and O.argus Cantor(♂)×O.maculatus(♀) reacted with six kinds of secondary antibodies(rabbit anti-chicken IgM,rabbit anti-pig IgG,rabbit anti-chicken IgG,rabbit anti-rat IgM,rabbit anti-monkey IgG and goat anti-human IgG)for Western blot.The serum immunnoglobulin level of T.sinensis and O.argus Cantor(♂)×O.maculatus(♀) was determined by using ELISA kits of chicken IgG,chicken IgM,swine IgG,rat IgM,mongkey IgG and human IgG.The serum immunnoglobulin concentrations of T.sinensis and O.argus Cantor(♂)×O.maculatus(♀) were 28.5-215.4 and 24.5-201.5 μg/L respectively.[Conclusion] This study could enrich the specific immune system research of T.sinensis and O.argus Cantor(♂)×O.maculatus(♀) and it was of certain guiding significance for the healthy breeding of T.sinensis and O.argus Cantor(♂)×O.maculatus(♀).
