陆地棉遗传背景下海岛棉Chromsome7片段纤维品质性状的QTL定位

【摘要】 为消除遗传背景的干扰,深入剖析陆地棉遗传背景下渗入的海岛棉Chromsome 7(Chr.7)片段对优异纤维性状的遗传贡献,本研究从(鲁棉研22×鲁原343)重组自交系中选择携带海岛棉Chr.7染色体片段的优质次级渐渗系R472作亲本,与高产亲本鲁棉研22回交,构建了包含514个F2单株的次级定位群体。利用JoinMap构建Chr.7的遗传图谱,运用基于混合线性模型的QTLNetwork-2.2软件和基于多元回归模型的Windows QTL Cartographer 2.5软件对纤维品质性状进行QTL定位,结果显示,利用Windows QTL Cartographer 2.5检测到1个纤维强力QTL(qFS-C7-1),距离渐渗位点DC40182 8.1 cM,可解释表型变异率5.66%,增效等位基因来源于携带有海岛棉Chr.7染色体渐渗片段的R472;利用QTLNetwork-2.2检测到1个纤维强力QTL(qFS-C7-1)、1个纤维细度QTL(qFM-C7-1)。2个作图软件定位的纤维强力QTL位点基本一致,可认为是同一位点。

【Abstract】 In order to analyze the genetic contribution of introgressive segments of chromosome 7( Chr.7) to fiber traits,a secondary mapping population including 514 individuals was developed from a cross between secondary introgression line( SIL) R472 containing chromosomal segment of Chr. 7 from Gossypium barbadense,and recurrent parent LMY22. The linkage map was constructed using JoinMap,and the QTL identification was performed by QTLNetwork- 2. 2 and composite interval mapping approach of Windows QTL Cartographer 2. 5. The results showed that one QTL for fiber strength( qFS- C7- 1) was detected by Windows QTL Cartographer 2. 5,which had the distance to the site of DC40182 as 8. 1 cM,and explaining 5. 66% of the phenotypic variance; the favorable alleles were derived from the R472 parent. One QTL for fiber strength( qFS- C7-1) and one for fiber fineness( qFM- C7-1) were detected by QTLNetwork-2. 2. The QTLs for fiber strength detected by the two mapping software were located in the same interval.
