EGFRvⅢ在人脑成胶质细胞瘤中的表达及其对成胶质细胞瘤A172细胞增殖的影响
The expression of EGFRvⅢ in human glioblastoma and its effect on the proliferation of A172 cells
【摘要】 目的:探讨表皮生长因子受体Ⅲ型突变体(epidermal growth factor receptor variantⅢ,EGFRvⅢ)在人脑成胶质细胞瘤中的表达及其对成胶质细胞瘤A172细胞增殖的影响。方法:应用免疫组织化学染色法检测EGFRvⅢ在60例人脑成胶质细胞瘤及其癌旁正常脑组织中的表达。建立稳定高表达EGFRvⅢ的成胶质细胞瘤A172细胞株,应用细胞计数法、MTT法、细胞克隆形成实验以及裸鼠成瘤实验检测其对成胶质细胞瘤A172细胞增殖的影响。结果:EGFRvⅢ在脑成胶质细胞瘤组织中的阳性表达率明显高于其癌旁正常脑组织(P<0.01);与转染空载体对照组比较,高表达EGFRvⅢ的A172细胞增殖能力明显增加;对照组和高表达EGFRvⅢ组的细胞克隆数分别为(50±10)个和(140±25)个,差异有统计学意义(P<0.01);高表达EGFRvⅢ组的裸鼠皮下移植瘤的体积和质量均高于对照组(P<0.01)。结论:EGFRvⅢ在人脑成胶质细胞瘤组织中高表达,并且可促进成胶质细胞瘤A172细胞的增殖。
【Abstract】 Objective: To investigate the expression of EGFRvⅢ (epidermal growth factor receptor variant Ⅲ) in human glioblastoma tissues and its effect on the proliferation of A172 cells. Methods: The expressions of EGFRvⅢ in 60 glioblastoma tissues and their corresponding para-cancerous normal brain tissues were detected by immunohistochemistry. A172 cells overexpressing EGFRvⅢ were established. The effects of EGFRvⅢ overexpression on the proliferation of A172 cells were detected by cell counting, MTT method and colony formation assay as well as tumorigenesis experiment in nude mice. Results: The positive rate of EGFRvⅢ expression in glioblastoma tissues was higher than that in the corresponding para-cancerous normal brain tissues (P < 0.01). The proliferation ability of A172 cells overexpressing EGFRvⅢ was increased compared with that of the cells transfected with empty vector (as a control). The numbers of colony formation in the control cells and the EGFRvⅢ-overexpressing cells were 50±10 and 140±25, respectively (P < 0.01). The volume and weight of EGFRvⅢ-overexpressing A172 xenografts in nude mice were higher than those of the control A172 xenografts (P < 0.01). Conclusion: The overexpression of EGFRvⅢ in human glioblastoma tissues promotes the proliferation of A172 cells.
