白芍总苷在免疫性肝损伤小鼠药代动力学研究

【摘要】 目的:研究白芍总苷在免疫性肝损伤小鼠体内的药动学特点。方法:采用卡介苗加脂多糖尾静脉注射法建立小鼠免疫性肝损伤模型,HPLC法测定模型小鼠和正常小鼠灌胃给予白芍总苷(0.47g/kg、1.41g/kg、2.82g/kg)后15、30、60、90、120、150、180、240、360、480、720min血浆中芍药苷和芍药内酯苷浓度,根据药时曲线计算药动学参数,采用SPSS11.5软件分析各组各剂量间药动学参数的异同。结果:与正常组相比,模型组小鼠体内白芍总苷的Tmax明显提前,t1/2明显延长,Cmax、AUC0-t和AUC0-∞显著增大;各剂量间白芍总苷的Tmax和t1/2没有差异,芍药苷剂量与Cmax、AUC0-t和AUC0-∞有一定的相关性。结论:免疫性肝损伤小鼠对白芍总苷的吸收速度较正常小鼠加快,吸收量较大,消除减慢。

【Abstract】 Objective: To study pharmacokinetics of total glucosides of paeony(TGP) in immunological liver injuried mice.Methods: BCG and LPS caudal intravenous injection was used to establish immunological liver injuried mice.TGP(0.47,1.41,2.82 g/kg) was orally administered to normal and model animals.With HPLC,the concentration of Paeoniflorin and Albiflorin in plasma at times of 15,30,60,90,120,150,180,240,360,480 and 720 min after dosing was determined.The parameters were calculated according to concentration-time curve.SPSS 11.5 was used to analyze dates between groups.Results: Compared with normal group,Tmax of TGP in model group significantly advanced,t1/2 prolonged,Cmax,AUC0-t and AUC0-∞ increased;between dosages,Tmax and t1/2 of TGP hadn’t difference,and dosage of Paeoniflorin with Cmax,AUC0-t and AUC0-∞ had determinate correlation.Conclusions: TGP in immunological liver injuried mice was absorbed more rapidly than normal mice,absorbed dose larger,and eliminated more slowly.
