天麻素对长春新碱诱导大鼠神经病理性疼痛的抑制作用
The inhibitive effects of gastrodin on the vincristine-induced neuropathic pain
【摘要】 目的:观察天麻素对长春新碱诱导大鼠神经病理性疼痛的影响。方法:用长春新碱125μg/kg隔日腹腔注射制作大鼠化疗痛动物模型,腹腔注射天麻素治疗;用电子Von Frey测痛仪测定大鼠机械痛阈值,电子压痛仪测定大鼠压痛痛阈值,电子热板仪测定大鼠热刺激痛阈值。结果:腹腔注射长春新碱在给药第10天与对照组比较三种刺激痛阈值均有显著性降低(P<0.01),化疗痛模型成功建立之后,加用天麻素腹腔注射1~2周,不同剂量天麻素组(30mg/kg、60mg/kg、120mg/kg)的三种刺激痛阈值与模型组比较均有不同程度的提高,且呈剂量依赖性,以大剂量天麻素的作用最明显(P<0.01),且天麻素治疗1周与治疗2周的痛阈值无差异。结论:天麻素对长春新碱诱导的大鼠痛觉过敏有抑制作用;持续给药该作用可以维持,有利于延长长春新碱抗肿瘤的疗程,以提高其抗肿瘤疗效。
【Abstract】 Objective: To observe effects of gastrodin on the vincristine-induced neuropathic pain in rat.Methods: Vincristine was administered intravenously on alternate days until five injections,after the model of chemotherapy-induced neuropathic pain was established successfully,Gastrodin was administered intravenously.Mechanical allodynia was measured through observing withdraw claw reaction of rat using electronic Von Frey.Tenderness hyperalgesia was measured in tail by pressing stimulator,heat hyperalgesia was measured by using electron ic heatingplaten instrument.Results:After administering vincristine ten days,compared with the control group,the three types pain threshold were decreased significantly(p<0.01).Gastrodin was injected in addition everyday,after vincristine-induced neuropathic pain model was established successfully,mechanical pain and press pain and heat pain threshold were different degree improved in different dose treatment groups compared with the model group.the effect of high dosage Gastrodin was most obvious(p<0.01).And the pain threshold wasn’t different after injecting Gastrodin one or two weeks.Conclusion: Gastrodin has inhibition action to vincristine-induced hyperalgia in rat and the action can be maintained when medicine was used persistently,the least 1 week.This can be helpful to prolonging the course of treatment of vincristine antineoplastic treatment and enhance its efficacy.
