刺五加对果蝇睡眠剥夺模型睡眠及全基因表达谱的影响

【摘要】 目的:探讨刺五加对睡眠剥夺果蝇睡眠的改善作用,并进行差异基因表达谱初步分析。方法:在恒温、恒湿、自动光控条件下,选取7日龄野生型Canton S品系黑腹果蝇为试验对象,采用光照刺激的方法复制果蝇睡眠剥夺模型,利用自动红外果蝇活动监测系统,以果蝇24h总睡眠时间和睡眠指数为观察指标,进行刺五加改善睡眠剥夺果蝇睡眠作用的量-效与时-效关系研究;在此基础上,采用果蝇表达谱芯片技术进行刺五加改善果蝇睡眠的差异基因表达谱初步分析。结果:刺五加浓度1%、给药3天以上,对果蝇睡眠的改善作用最明显;与模型组相比,刺五加组共有差异表达基因685个,上调基因188个,下调基因497个。结论:刺五加对果蝇睡眠剥夺模型的睡眠具有明显的改善作用,其作用机制涉及免疫反应和多细胞生物过程等相关基因表达的变化。

【Abstract】 Objective:To study the effects of Acanthopanax senticosus on sleep and gene expression profiles of sleep-deprivated drosophila.Methods:Under the standard conditions of costant temperature,humidity,automatically controlled lighting,an infrared drosophila activity monitor system(DAMS) was used to monitor the sleeping data of drosophila,and the dose-effect and time-effect of Acanthopanax senticosus on improving sleep was studied in 7-day-old drosophila;then,effects of Acanthopanax senticosus on gene expression profiles of sleep-deprivated drosophila were studied by the microarray technology.Results:1% drug medium for dose,≥3days for delivery time is best for improving sleep of drsophila;Acanthopanax senticosus(concentration of 1%,administration 4 days) can changed expression of 685 genes in the head of fly,188 genes were up-regulated,and 497 genes were down-regulated.Conclusions:Acanthopanax senticosus can improving sleep of drosophila,and the mechasisms involved in the process of the immune response and multicellular organism.
