吴茱萸水提组分单次给药对小鼠肝毒性“量-时-毒”关系研究
“Dose-time-toxicity” relationship study on hepatotoxicity caused by single dose water extraction components of Evodia Fructus to Mice
【摘要】 目的:研究吴茱萸水提组分单次给药致小鼠肝毒性的时毒、量毒关系。方法:取小鼠按不同时间点和不同剂量分组给药,观察小鼠死亡情况和毒性反应,检查血清谷丙转氨酶(ALT)、谷草转氨酶(AST),计算肝、脾、胸腺脏器指数,光学显微镜下观察肝组织形态变化。结果:"时-毒"关系研究表明:小鼠单次灌服32.5 g/kg的吴茱萸水提组分后,血清ALT、AST值随时间不同均造成肝损害程度也不同,均可导致肝脏指数明显升高,小鼠血清ALT、AST均在给药后2 h达到高峰;肝组织病理学检查显示,吴茱萸水提组分可在给药8~72 h内对肝组织产生明显损伤。"量-毒"关系研究表明:与正常组比较,吴茱萸水提组分不同剂量组对肝组织产生不同程度的损伤,且随着剂量增大,ALT、AST升高显著,呈剂量依赖关系。结论:小鼠单次灌服吴茱萸水提组分可造成小鼠一定的急性肝损伤,其对肝损伤程度呈现有一定的"量-时-毒"关系。
【Abstract】 Objective:To study on the time-toxicity and dose-toxicity relationships caused by single dose water extraction components of Evodia Fructus to mice.Methods:Mice were packeted according to different time and dose,/ to observe the death circumstance and toxicity of mice,detect ALT 、AST in serum,calculate liver,spleen,thymus index,and observe the morphological changes of liver tissue under light microscope.Results:The "time-toxicity" relationship showed that:ALT,AST in mice serum administrated to 32.5 g/kg water extraction components of Evodia Fructus were changed differently in different time,the liver index had significant ly increased,and ALT,AST in mice serum were peaked after 2 hours’ administration;could produce significant liver damage.The "dose-toxicity" relationship showed that:compared the histopathological examination of of hepatic tissue showed that the water extraction components of Evodia Fructus after 8~72 hours’ administration to mice with the normal group,the water extraction components of Evodia Fructus which had different dose could produce different degree’s damage to hepatic tissue,the ALT,AST in serum increased significantly with the dose increased and have dose-dependence ralationship.Conclusion:Single dose water extraction components of Evodia Fructus to mice could produce acute liver injury,and there were certain time-toxicity and dose-toxicity relationships.
