【摘要】 目的:研究朱砂七总蒽醌(ZSQ)对HL-60细胞增殖抑制和诱导凋亡的作用,探讨其抗肿瘤作用及其机制。方法:应用MTT法检测朱砂七总蒽醌对HL-60细胞的生长抑制作用,Giemsa染色观察细胞形态学改变,DNA ladder和流式细胞术等方法检测朱砂七总蒽醌诱导肿瘤细胞凋亡及对细胞周期的作用。结果:32、3.2、0.32、0.032mg/ml朱砂七总蒽醌作用HL-60细胞72h后,细胞生长抑制率分别为28.69%,36.84%,63.76%和80.57%,抑制作用呈时间及浓度依赖性,朱砂七总蒽醌在48h的IC50约为3.2mg/ml。形态学观察到用药后凋亡细胞典型的形态学特征,伴有多核细胞形成。FCM分析发现朱砂七总蒽醌阻断HL-60细胞生长于细胞周期的G2/M期,诱导凋亡作用呈时间及浓度依赖性,能够诱发肿瘤细胞产生凋亡小体、DNA ladder和细胞凋亡峰。结论:朱砂七总蒽醌能抑制HL-60肿瘤细胞的增殖并诱导凋亡。更多还原

【Abstract】 Objective:To investigate the effects and related mechanism Of ZSQ on the proliferation inhibition and induction of apoptosis in HL-60 cell.Methods:We investigated the action ZSQ against the survival of cancer cell line HL-60 with MTT assay.morphologic changes of cell were observed by Giemsa staining.DNA gel electrophoresis and flow cytometry analysis were used to determine tumor cell apoptosis and cell cycle induced by ZSQ.Results:HL-60 cells were treated with 32、3.2、0.32 and 0.032mg/ml of ZSQ for 72 h,the inhibitory rate cell growth was 28.69%,36.84%,63.76% and 80.57%,respectively.ZSQ have a time and dose-dependent growth inhibition.The IC50 was 3.2mg/ml.After tumor cells were treated with ZSQ,Morphological characteristic of apoptosis was detected,A majority of HL-60 cell were arrested in G2-M phaseand,Agarose gelelectrophoresis showed typical DNA ladder.Flow cytometric analysis showed apoptotic"Sub-Gl peak".Conclusion:ZSQ can inhibit the proliferation of tumor cells and induces apoptosis in HL-60.更多还原
