解毒活血法对单侧输尿管梗阻大鼠PRA和AngⅡ的影响

【摘要】 目的:观察解毒活血法对单侧输尿管梗阻大鼠PRA、AngⅡ的影响,探讨其抑制肾间质纤维化的作用机制。方法:SD大鼠随机分为假手术组、模型组、缬沙坦组、解毒活血低剂量组、解毒活血高剂量组,每组10只。除假手术组其余各组大鼠结扎左侧输尿管复制UUO模型,所有大鼠灌胃给药,缬沙坦组给予26mg/kg·d,解毒活血低剂量组给予解毒活血中药12g生药/kg·d,解毒活血高剂量组给予24g/kg·d,假手术组和模型组给予等容量生理盐水共14d。取左肾组织行HE、Masson染色观察病理形态学改变,用放射免疫法检测血及组织中的PRA、AngⅡ表达。结果:模型组大鼠血清及肾组织中PRA、AngⅡ水平明显升高(P<0.05)。经治疗各组大鼠血清及肾组织中PRA、AngⅡ水平显著下降(P<0.05)。结论:解毒活血方药能够降低PRA、AngⅡ水平,阻断RAS,保护肾功能,延缓肾间质纤维化进展。

【Abstract】 Objective:To observe the effects of Jiedu Huoxue therapy on the expression of PRA、AngⅡ in UUO rats,and to investigate its mechanism in tubulointerstitial fibrosis.Methods: Fifty Sprague-Dawley rats were randomized equally into 5 groups: Control group,UUO group,Valsartan group,low-dose of Jiedu Huoxue decoction group and high-dose of Jiedu Huoxue decoction group.Except the Control group,rats in other four groups were made into UUO model by ligaturing the left ureter.All rats were treated with drugs by oral infusion once a day.Rats in Valsartan group,Jiedu Huoxue decoction low-dose group and high-dose group were treated respectively with Valsartan 26mg/kg·d,Jiedu Huoxue decoction 12g/kg·d and 24g/kg·d for fourteen days.Take the left kidney to detect PRA and AngⅡby radioimmunoassay.Results: The expression of PRA and AngⅡ in tissue and serum were increased significantly in the model group(P<0.05).After treatment,the expression of PRA and AngⅡ were significantly down-regulated(P<0.05).Conclusions: Jiedu Huoxue decoction plays a role in down-regulating the over expression of PRA、AngⅡ,so as to retard the progress of renal interstitial fibrosis through inhibiting RAS.
