柴胡疏肝散对阿尔茨海默病大鼠海马CA1区超微结构的影响
Effects of Chaihu Shugan San on ultrastructure of hippocampal CA1 in Alzheimer’s Disease rats
【摘要】 目的:探讨柴胡疏肝散在阿尔茨海默病(Alzheimer’s diseas,AD)治疗中的意义。方法:大鼠随机分为6组,其中模型组(C),柴胡疏肝散低剂量和高剂量组(D、E),补肾组(F)均采用D-半乳糖和Aβ双侧海马注射联合造模,Aβ造模成功后的第9天,D、E组分别ig给予柴胡疏肝散10,20 g.kg-1,F组给予脑力宝20 g.kg-1。各组均在每天上午9∶00按10 mL.kg-1的容积ig给药,连续ig28 d。ig前后分别进行水迷宫行为学检测。每组取3只,在ig干预结束后的第4天处死动物取海马,Leica EMUC-7超薄切片,透射电镜观察海马CA1区超微结构。结果:柴胡疏肝散高剂量组ig前后迷宫寻找平台时间分别为17.34±3.35s,12.78±2.54s,补肾组分别为17.79±2.25s、14.11±2.66s,2组ig前后比较及ig后与同期的模型组比较均有显著差异(P<0.01)。电镜显示,疏肝低剂量组神经元结构基本正常,突触前膜内突触小泡较清晰,突触间隙清晰,线粒体膜基本完整,线粒体嵴较清晰。疏肝高剂量组神经元结构正常,突触小泡及突触间隙清晰,线粒体膜完整,线粒体嵴清晰可见;而模型组电镜下神经细胞形态破坏,结构紊乱,突触密度降低且均匀不一,突触间隙变小不清,突触小泡数量减少,游离核蛋白体减少,线粒体膜不完整,线粒体嵴模糊。结论:大剂量柴胡疏肝散对AD大鼠模型脑细胞有一定的保护作用,在AD治疗中应重视疏肝理气法的应用。
【Abstract】 Objective: To investigate the Chaihushugansan on Alzheimer s diseas(AD) and treatment of.Methods: the rats were randomly divided into six groups,in which the model group(C),Chaihu Shugan powder of low dose and high dose group(D,E),Bushen group(F) using D-galactose and Aβ bilateral hippocampal injection molding,A beta model after the success of the ninth D,D,E group were given Ig Chaihu Shugan powder 10,20 g·kg-1,F group received mental treasure 20 g·kg-1.All groups were in the morning of day 9∶00 according to the 10 mL·kg-1 Ig volume delivery,continuous ig28 D.Ig were performed before and after the water maze behavior detection.Each group of 3 only,the Ig intervention after the end of the fourth d to take Leica EMUC-7 animal hippocampus,ultrathin section,transmission electron microscopic study on the ultrastructure of hippocampus CA1 area.Results: the high dose group of Chaihu Shugan powder before and after ig maze to find the platform respectively 17.34±3.35s、12.78±2.54s,Bushen group were 17.79±2.25s、14.11±2.66s,two groups of Ig and Ig before and after comparison with earlier than in model group were significantly different(P<0.01).Electron microscopy showed low dose group of neurons on the normal construction,presynaptic membrane of synaptic vesicles is clear,the synaptic cleft is clear,mitochondrial membrane integrity,mitochondrial cristae clearer.Soothing the liver and the high dose group,normal neuronal structure,synaptic vesicles and synaptic gap is clear,mitochondrial membrane integrity,mitochondrial crest clearly visible.While the model group under electron microscope of nerve cell morphology damage,structural disorder,decreased synaptic density,and uneven,the synaptic gap smaller unclear,reduced quantity of synaptic vesicles,free ribosomes decrease,mitochondrial membrane integrity,mitochondrial cristae vague.Conclusion: the large dosage of Chaihu Shugan powder on AD rat model brain cells have a protective effect,in AD treatment should be paid attention to in the application of the method of soothing the liver and regulating qi.
