【摘要】 目的:通过应用逆转录聚合酶链式反应(RT-PCR)技术分析内源性Ⅱb型MHC、Ⅰ型及Ⅲ型胶原mRNA的表达来探讨活血止痛膏剂对骨骼肌钝挫伤修复的作用及其可能机制。方法:用63只大白兔随机分为活血止痛膏高剂量组、中剂量组、低剂量组、青鹏膏对照组、外伤对照组,每组12只,另取3只做正常对照组。采用"重物自由落体打击法"制作大白兔骨骼肌急性钝挫伤模型。分别在3天、10天、20天、27天处死大白兔,每组每个时间点处死3只兔子,每只兔子在两条后腿腓肠肌取标本,共计6个标本。结果:(1)RT-PCR技术检测内源性Ⅱb型MHC mRNA,第3、10、20天高、中剂量组和青鹏膏组内源性Ⅱb型MHC mRNA表达量高于外伤组、低剂量组(P<0.05);第10、20天高剂量组对内源性Ⅱb型MHC mRNA表达高于中剂量组(P<0.05);(2)RT-PCR技术检测内源性Ⅰ型胶原mRNA的表达量:第3天各组比较没有统计学意义;第10、20天高、中剂量组和青鹏膏组表达量低于外伤组、低剂量组(P<0.05);第10、20天高剂量组优于中剂量组(P<0.05)。(3)RT-PCR技术检测内源性Ⅲ型胶原mRNA的表达量:第3、10、20天高、中剂量组和青鹏膏组内源性Ⅲ型胶原mRNA表达低于外伤组、低剂量组(P<0.05);第3、10、20天高剂量组和青鹏膏组对内源性Ⅲ型胶原mRNA表达量明显少于中剂量组(P<0.05)。3个检测指标各个时间点青鹏膏组和高剂量组比较无统计学意义(P>0.05),即作用效果没有差别。结论:(1)活血止痛膏可有效的促进骨骼肌急性钝挫伤后Ⅱb型MHC的mRNA表达的提高。(2)活血止痛膏能促进骨骼肌急性钝挫伤后Ⅰ、Ⅲ型胶原mRNA表达的减少。(3)活血止痛膏可通过促进Ⅱb型MHC mR-NA的表达,同时减少Ⅰ、Ⅲ型胶原mRNA表达提高骨骼肌愈合质量。(4)高剂量活血止痛膏与青鹏膏在骨骼肌Ⅱb型MHC mRNA表达和Ⅰ、Ⅲ型胶原mRNA表达效果相当。更多还原

【Abstract】 Objective:To investigate the role and possible mechanism of Huoxue Zhitong cream repairing muscle contusion by Reverse Transcription Polymerase Chain Reaction(RT-PCR)technique and to analyze the expression of endogenous type Ⅱb MHC mRNA,Ⅰ and Ⅲ collagen mRNA.Method:63 rabbits were randomly divided into Huoxue Zhitong cream high dose group,middle dose group,low dose group,Qingpeng Ointment group,trauma control group.Each group contained 12 rabbits,and there were 3 rabbits as normal control.The method of Free-falling weights was used to make the model with acute rabbit’s skeletal muscle contusion.The rabbits were killed on the 3rd,10th,20thand 27th day respectively,3 rabbits were killed at each time point in each group,specimens were taken on the gastrocnemius of the legs.Results:(1)The expression amount of endogenous type Ⅱb MHC mRNA by RT-PCR technique on the 3rd,10th and 20th day in the high dose,middle dose and Qingpeng Ointment groups was higher than trauma control group and low dose group.On the 10th and 20th day,that in the high dose group was higher than the middle dose group.(2)The expression amount of endogenous type I collagen mRNA RT-PCR technique on the 3rd day had no statistical significance among groups;on the 10th and 20th day,that in the high dose,middle dose and Qingpeng Ointment groups was lower than that in trauma control group and low dose group while that in the high dose group was higher than that in the middle dose group.(3)The expression amount of endogenous type Ⅲ collagen mRNA by RT-PCR technique on the 3rd,10th and 20th day in the high dose,middle dose and Qingpeng Ointment groups was lower than that in trauma control group and that in low dose,high dose and Qingpeng Ointment groups was lower than that in the middle dose group.Of the three marks,there was no statistical significance between the Qingpeng Ointment group and the high dose group at each time point.Conclusion:First,Huoxue Zhitong cream can significantly promote the expression of IIb-type MHC mRNA after acute contusion of rabbit skeletal muscle.Second,Huoxue Zhitong cream can reduce the expression of Ⅰ,Ⅲ collagen mRNA after acute contusion of rabbit skeletal muscle.Third,Huoxue Zhitong cream can promote the expression of IIb-type MHC mRNA and reduce the expression of Ⅰ,Ⅲ collagen mRNA,thus it can improve the healing quality of skeletal muscle.Fourth,the expression of type IIb MHC mRNA and Ⅰ,Ⅲ collagen mRNA of skeletal muscle has comparable effect between the high dose group and the Qingpeng Ointment group.更多还原
