铝暴露对大鼠海马的影响及其机制研究
Influence of Exposure to Aluminium on Rat Hippocampus And Its Mechanism
【摘要】 目的:通过观察铝暴露后大鼠记忆行为学、电生理长时程增强(LTP)、神经生化(ERK含量)变化,探讨铝物质对学习记忆的影响作用机制,并为中医理论"脑主神明"提供可靠的实验依据。方法:实施铝暴露:给大鼠自由饮用不同剂量AlCl3溶液。观察鼠Morris水迷宫行为学表现、细胞外微电极记录各组海马DG-CA3途径的LTP(长时程增强)、蛋白免疫印迹(Western-Blot)方法测定细胞外信号调节激酶(ERKs)活性。结果:对照组、低剂量组、高剂量组的行为学表现、LTP、Western-Blot ERKs表达比较有显著差异(P<0.05)。结论:铝在体内蓄积可以影响大鼠的学习记忆,并为中医理论"脑主神明"提供实验依据。
【Abstract】 Objective:Through the observation of learning and memory,electrophysiological long-term potentiation(LTP),biological and chemical nerve(ERK content) after exposure to aluminum,discussion of aluminum material impact on learning and memory mechanism was made,providing a reliable basis for TCM theory of brain governing mind.Methods:The implementation of aluminum exposure:rats were given different doses AlCl3 solution for free drinking.Observe rats behavioral performance in water maze,extracellular microelectrode was used to record the the LTP of DG-CA3 way,Western-Bloting determined ERKs activity expression in each group.Results:Behavioral performance,LTP and Western-Blot ERKs expression in control group,low dose group and high dose group were significantly different(P<0.05).
