岩大戟内酯B诱导乳腺癌MCF-7细胞凋亡的实验研究
The Experiment Research on Apoptosis of Breast Cancer Cell Line MCF-7 Treatment with Jolkinolide B
【摘要】 目的:探讨岩大戟内酯B对MCF-7乳腺癌细胞生长抑制作用及其机理。方法:通过MTT法观察各组细胞增殖能力;采用流式检测技术分析细胞周期;利用AnnexinV/PI双染检测细胞凋亡情况;电镜观察细胞形态。结果:随着岩大戟内酯B浓度增加对MCF-7的抑瘤率增高;G2/M期细胞减少,S期细胞增多,出现S期阻滞;晚期凋亡和死亡的细胞增加;电镜显示加药后细胞出现核固缩,细胞器空泡变性,凋亡小体形成,甚至出现坏死。结论:岩大戟内酯B对乳腺癌MCF-7细胞生长明显抑制作用,并诱导肿瘤细胞凋亡,为临床开发新药提供理论依据。
【Abstract】 Objective:To investigate the inhibition effects of Jolkinolide B on breast cancer cell growth inhibition and its mechanism.Methods:Author used MTT method to observe the proliferation of MCF-7 cell lines when treat with Jolkinolide B;Cell cycle was assayed by flow cytometer;Annexin V/PI double dye was detected to cell apoptosis;Cell morphology was observed by electronic microscopy.Results:The inhibit ratios of MCF-7 cell lines were increased following the drug concentrations of Jolkinolide B;The G2 /M period cells reduce,S phase increased the number of cells and increase apoptosis rates,it appears the block of S phase;There were a lot of apoptotic cell and dead cell;Many nuclei had become pyknotic by electronic microscopy,organelles empty bubble degeneration,appear apoptotic body,even necrosis after dosing the different concentration of drug.Conclusion:Jolkinolide B can significantly inhibit the growth of MCF-7 breast cancer cell and induced apoptosis.It is provides the theory for a new drug with the development of clinical therapy.
