葱白提取物和丹参酮ⅡA磺酸钠对急性心肌缺血兔血清CRP、SOD、MDA水平影响的实验研究
Experimental Study on the Influence of Fistular Onion Stalk Extract and Sodium Tanshinone ⅡA Sulfonate on Serum Levels of CRP,SOD and MDA in Acute Myocardial Ischemic Rabbit
【摘要】 目的观察葱白提取物和丹参酮ⅡA磺酸钠对急性心肌缺血兔血清C-反应蛋白(CRP)、丙二醛(MDA)和超氧化物歧化酶(SOD)的影响,探讨葱白提取物和丹参酮ⅡA磺酸钠治疗急性心肌缺血的作用机制并比较其优势。方法选用体重为2.5~3.0kg的健康雄性日本大耳白兔40只,随机分为假手术组、空白模型组、丹参酮ⅡA磺酸钠、硝酸甘油组和葱白提取物组,每组8只。制备急性心肌缺血模型,术后及时给药,假手术组和模型组舌下给予生理盐水(0.2ml/kg),葱白提取物组舌下给予葱白提取物混悬液(20mg/kg、100mg/ml),硝酸甘油组舌下给予硝酸甘油混悬液(1mg/kg、5mg/ml),丹参酮ⅡA磺酸钠组舌下给予丹参酮ⅡA磺酸钠(1mg/kg、5mg/ml)。给药后观察3h,打开腹腔,腹主动脉取血,测定血清CRP、SOD和MDA含量。结果与模型组比较,葱白提取物组、丹参酮ⅡA磺酸钠组、硝酸甘油组血清中MDA水平均显著降低(P<0.05),而SOD水平均明显升高(P<0.01)。葱白提取物组和丹参酮ⅡA磺酸钠组SOD水平均明显高于硝酸甘油组(P<0.05),葱白提取物组和丹参酮ⅡA磺酸钠组之间SOD水平无统计学差异。葱白提取物组、丹参酮ⅡA磺酸钠组、硝酸甘油组血清中MDA水平比较差异无统计学意义(P>0.05)。葱白提取物组和丹参酮ⅡA磺酸钠组血清CRP水平的降低差异无统计学意义(P>0.05),均明显优于硝酸甘油组(P<0.01)。结论葱白提取物和丹参酮ⅡA磺酸钠均可降低MDA和CRP水平,升高SOD水平,两者作用差异无统计学意义(P>0.05),表明通阳中药葱白有效部位提取物和活血化瘀中药丹参有效成分丹参酮ⅡA磺酸钠均可以清除氧自由基,抗氧化损伤。
【Abstract】 Objective To observe the impact of fistular onion stalk extract and sodium tanshinone ⅡA sulfonate on serum C-reactive protein(CRP),malondialdehyde(MDA)and superoxide dismutase(SOD)in acute myocardial ischemic rabbit,to probe the mechanism of fistular onion stalk extract and sodium tanshinone ⅡA sulfonate in the treatment of acute myocardial ischemia,and to compare their benefits.Methods Forty health male Japanese white rabbits with the body weight of 2.5-3.0 kg were selected and randomly divided into sham operation group,the blank model group,sodium tanshinone ⅡA sulfonate group,nitroglycerin group and fistular onion stalk extract group(n=8).As soon as the model rabbits with acute myocardial ischemia were prepared,they were given different interventions.The sham operation group and model group were given saline(0.2 ml/kg)sublingually.The fistular onion stalk extract group was give light blue extract suspension(20 mg/kg,100 mg/ml)sublingually.The nitroglycerin group was given nitroglycerin suspension(1 mg/kg,5 mg/ml)sublingually.The sodium tanshinone ⅡA sulfonate group was administered sodium tanshinone ⅡA sulfonate(1 mg/kg,5 mg/ml)sublingually.Three hours later,the abdominal cavity was open and abdominal aortic blood was collected.Serum levels of CRP,SOD and MDA were determined.Results Compared with the model group,serum MDA levels in fistular onion stalk extract group,sodium tanshinone ⅡA sulfonate group and nitroglycerin group reduced significantly(P<0.05),whereas SOD levels increased obviously(P<0.01).SOD levels in fistular onion stalk extract group and sodium tanshinone ⅡA sulfonate group increased more obviously than that in nitroglycerin group(P<0.05),without difference in SOD levels between fistular onion stalk extract group and sodium tanshinone ⅡA sulfonate group.There was no difference in MDA levels among fistular onion stalk extract group,sodium tanshinone ⅡA sulfonate group and nitroglycerin group(P>0.05).Serum CRP levels in fistular onion stalk extract group and sodium tanshinone ⅡA sulfonate group reduced more significantly than that of the nitroglycerin group(P<0.01).There was no significant difference(P>0.05)between the serum CRP levels in fistular onion stalk extract group and sodium tanshinone ⅡA sulfonate group.Conclusion Both fistular onion stalk extract and sodium tanshinone ⅡA sodium can reduce the MDA and CRP levels and increase SOD levels,without significant difference between their effects,which indicated that both fistular onion stalk extract(effective extract of light blue)and sodium tanshinone ⅡA sulfonate sodium(active ingredients of traditional Chinese medicine Danshen)could scavenge oxygen free radicals and counteract oxidative damage.
