TRAIL基因多态性与甲状腺疾病的临床实验研究
Clinical study about polymorphism of TRAIL gene and Thyroid Disease
【摘要】 目的探讨细胞凋亡因子(TRAIL)1525位点基因多态性在中国汉族包头地区人群中的分布及其与甲状腺疾病的相关性。方法选择272例均参照2008年《中国甲状腺疾病诊治指南》确诊甲状腺疾病患者和100例正常对照组作为研究对象,应用聚合酶链式反应-限制性内切酶片段长度多态性(PCR-RFLP)方法进行多态性检测。结果结节性甲状腺肿组、腺瘤组与对照组TRAIL基因1525位点多态性比较均具有显著性差异(P<0.05);但结节的大小在各基因型间的比较,差异无显著性(P>0.05)。按性别分层后发现,各实验组女性患者与对照组女性间TRAIL基因1525位点多态性比较均具有显著性差异(P<0.05)。Graves病组中合并突眼与否及桥本甲亢与桥本甲减组TRAIL基因多态性分布组间比较,差异无显著性(P>0.05)。结论 TRAIL基因1525位点多态性与甲状腺疾病明显相关,提示TRAIL基因可能参与了甲状腺疾病的病理生理过程。
【Abstract】 【Objective】 Probe in TRAIL +1525 G/A gene polymorphism in patients with thyroid disease;Systematical research on the relation between individual gene polymorphism and thyroid diseases in Chinese patients of Baotou area.【Methods】 272 patients with thyroid disease diagonosed according to 2008 year about guidance of thyroid disease for treatment and diagnose in china and 100 healthy control subjects,TRAIL gene 1525 polymorphism was typed by(PCR-RFLP).Alleles and genotypes were assessed by chi-squaretest,and measurement data of interclass by analyzed with variance test.【Results】 The comparison of the genotype distributions among nodular goiter group,adenoma group and healthy control were significantly different(P <0.05);But the relationship of nodule size with distribution of genotype Frequencies was not statistically significant(P >0.05).Stratified by gender and found that TRAIL +1525 G/A locus genotype frequency distribution among female patients of each experimental groups and healthy women’ group were significantly different(P <0.05).In Graves disease,TRAIL genotype and allele frequency distribution between groups combined with exophthalmos or not,the difference was not statistically significant(P >0.05).These results are equally applicable to Hashimoto’s thyroiditis group between hypothyroidism and hyperthyroidism(P >0.05).【Conclusion】 TRAIL gene 1525 polymorphism was significantly associated with thyroid disease,suggesting that TRAIL gene may be involved in the pathophysiology of thyroid disease.
