姜黄素抑制气道重塑小鼠平滑肌细胞增殖的作用

【摘要】 目的探讨姜黄素对气道重塑小鼠气道平滑肌细胞增殖的作用。方法 BALB/c小鼠随机分为对照组、模型组、姜黄素小剂量组、姜黄素中剂量组和姜黄素高剂量组。以腹腔注射卵白蛋白致敏和雾化激发建立慢性哮喘小鼠气道重塑模型。姜黄素组在每次雾化前30 min给予姜黄素干预,其他组给予生理盐水。观察小鼠激发时的症状。检测气道反应性。计数支气管肺泡灌洗液中嗜酸性粒细胞占白细胞总数的百分比。检测血清特异性IgE的含量。免疫组织化学鉴定气道平滑肌,图像分析软件IPP测支气管壁厚度、平滑肌层厚度、平滑肌细胞数以及胞外信号调节激酶(extracellular signal regulated kinase,ER K)在气道平滑肌细胞中的灰度值。结果与对照组相比,模型小鼠激发早期出现烦躁不安、呼吸急促和腹肌痉挛等阳性反应。姜黄素干预组症状不明显,出现症状较晚,且很快缓解。姜黄素干预组的气道反应性低于气道重塑模型组(P<0.05)。姜黄素干预组的支气管肺泡灌洗液中嗜酸性粒细胞占白细胞总数的百分比低于模型组(P<0.05)。姜黄素干预组的血清特异性IgE低于模型组(P<0.05)。姜黄素干预组的支气管壁厚度、支气管平滑肌层厚度及支气管平滑肌细胞数量均低于模型组(P<0.05)。结论姜黄素可以抑制哮喘小鼠气道重塑模型中气道平滑肌的增殖,其机制与姜黄素抑制ER K信号通路和减少嗜酸性粒细胞在肺组织中的浸润相关。

【Abstract】 【Objective】 To investigate inhibitory effect of curcumin on proliferation of airway smooth muscle cells in airway remodeling mice.【Methods】 The BALB/c mice were divided into control group,model group,low-dose curcumin group,middle-dose curcumin group and high-dose curcumin group.The mice were sensitized and challenged with ovalbumin to establish a mouse airway remodeling model.The mice of curcumin groups were given curcumin 30 minutes before stimulation,while the others given normal saline.Activity condition of the mice was observed closely when challenged,the airway responsiveness was detected,the percentage of eosinophils in total Leukocyte in the bronchoalveolar lavage fluid was measured and the content of ovalbumin special immunoglobulin E(IgE) in the sera were tested by ELISA.The airway smooth muscle cells were identificated by immunohistochemistry.Thickness of airway wall,thickness of airway smooth muscle layer,number of smooth muscles and the level of ERK were measured by IPP in the slides.【Results】 The mice of model group presented positive symptoms such as dysphoria,tachypnea and abdominal spasm in the earlier stage while the mice of curcumin group presented less symptoms later and relieved soon.The mice of control group didn’t present any symptom.The airway responsiveness of the mice of curcumin group was lower than that of the mice of model group(P <0.05).The percentage of eosinophils in total Leukocyte in the bronchoalveolar lavage fluid of the mice of curcumin group was less than that of the mice of model group(P <0.05).The content ovalbumin specific IgE in the sera of the mice of curcumin group was less than that of the mice of model group(P <0.05).The thickness of airway wall,the thickness of airway smooth muscle layer and the airway smooth muscle cells of the mice of curcumin group were less than those of the mice of model group(P <0.05).【Conclusion】 The proliferation of airway smooth muscle cells in the airway remodeling was inhibited by curcumin,and this may be related to inhibition of ERK signaling pathway and inhibition of eosinophils infiltrating in the lung.
