Bmi-1和hTERT与乳腺癌发生发展的关系

【摘要】 目的探讨Bmi-1、hTERT mRNA和蛋白在人乳腺癌发生发展中的作用及其相互之间存在的关系。方法采用免疫组化方法及RT-PCR技术,分别检测56例乳腺浸润性导管癌、35例乳腺纤维腺瘤及30例正常乳腺组织中Bmi-1、hTERT mRNA及蛋白的表达情况。结果 Bmi-1、hTERT mRNA和蛋白在乳腺癌组织中的表达明显高于其在乳腺纤维腺瘤及正常乳腺组织中的表达,三组间比较存在显著性差异(P<0.05)。Bmi-1和hTERT在乳腺癌中的表达呈显著性正相关(P<0.05)。结论 Bmi-1、hTERT mRNA及蛋白高表达与乳腺癌的恶变演进有关。Bmi-1基因调控hTERT基因的表达,在乳腺癌的发病过程中可能有重要意义。Bmi-1、hTERT表达水平的联合检测有助于乳腺癌的早期诊断及鉴别诊断,值得进一步研究。

【Abstract】 Objective To investigate the relationship between Bmi-1,hTERT mRNA,or protein expressions in the occurrence and development of breast carcinoma.Methods Human breast tissue samples were obtained from 56 invasive ductal carcinoma cases,35 breast fibroadenoma cases,and 30 normal breast tissue cases.The expressions of mRNA,protein of Bmi-1 and hTERT were examined by using immunohistochemistry(IHC) and RT-PCR,respectively.Results Both Bmi-1 and hTERT mRNA or protein expressions were significantly higher in breast invasive ductal carcinoma than those in normal breast tissue and breast fibroadenoma(P<0.05).Moreover,there was a significant positive correlation between Bmi-1 and hTERT expression in breast carcinoma(P<0.05).Conclusion A high Bmi-1 and hTERT mRNA or protein expressions could be considered associated with evolution and malignant transformation of the breast tumor.And Bmi-1 may play a role in the transcriptive activation of hTERT gene in breast carcinoma,which maybe has an effect on progression of breast carcinoma.Combined detection of Bmi-1 and hTERT expression may be useful for differential diagnosis of malignant and benign breast lesions,which deserves further researches.
