苦参碱对小鼠坐骨神经损伤后脊髓运动神经元的影响

【摘要】 目的探讨BALB/c小鼠坐骨神经损伤后应用苦参碱后对坐骨神经相应脊髓节段中gap-43的表达变化。方法健康成年BALB/c小鼠320只,单侧坐骨神经切断吻合后,随机分组给药,不同时间获取L4-6脊髓节段,用Real time-PCR检测坐骨神经相应脊髓节段gap-43的表达变化,同时进行髓鞘LFB染色观察坐骨神经的恢复情况。结果 Real time-PCR结果显示高剂量和中剂量组12h,24h3d5d1w,2w,4w,gap-43表达量明显高于低剂量组和空白对照组,8w各组比较无明显差异。LFB染色表明高中剂量组坐骨神经恢复情况明显优于低剂量组和对照组。结论坐骨神经损伤后应用苦参碱对坐骨神经脊髓相应节段细胞中gap-43的活化起到促进作用,减少炎症细胞的浸润从而达到促进神经再生的目的。

【Abstract】 Objective To investigate the BALB / c mice after sciatic nerve injury in the sciatic nerve after application of matrine corresponding spinal segment in the expression of gap-43.Methods Healthy adult BALB / c mice 320,consistent with unilateral sciatic nerve cut,the randomized dose,at different times for L4-6 spinal segment,using Real time-PCR detection of the corresponding spinal segment of sciatic nerve gap-43 expression changes,while the LFB myelin staining of sciatic nerve recovery.Results Realtime-PCR results indicate that high-dose and medium-dose group 12h,24h 3d 5d 1w,2w,4w,gap-43 expression was significantly higher than low-dose group and blank control group,8w no significant difference between the groups.LFB staining showed that the recovery of the sciatic nerve high-dose group was significantly better than low-dose group and control group.Conclusion application of sciatic nerve injury in the spinal cord of matrine on the corresponding segments of sciatic nerve cells in the activation of gap-43 play a catalytic role in reducing the infiltration of inflammatory cells to achieve the purpose of promoting nerve regeneration.
