【摘要】 目的比较福尔马林固定石蜡包埋组织(FFPET)中3种DNA提取方法,研究哪种方法更简便,更适合做DNA甲基化特异性PCR(MSP)。方法分别用二甲苯脱蜡法,改良TES水浴法和试剂盒法提取乳腺癌组织中的DNA,PCR扩增进行比较。结果二甲苯脱蜡法和试剂盒法提取的DNA电泳条带清晰可见,改良TES脱蜡法未见条带。结论二甲苯脱蜡法和试剂盒法提取的DNA均可用于做甲基化特异性PCR。更多还原

【Abstract】 Objective To evaluate which methods in DNA extraction from formalin-fixed,paraffin-embedded tissues is more simple and reliable for DNA methylation specific PCR(MSP).Methods Three methods,including xylene dewaxing method,improved TES dewaxing with water-bath method and DNA extraction kit method,were performed to extract DNA from breast cancer tissues and then conducted PCR amplification.Results The quality of DNA was high in xylene dewaxing method and in DNA extraction kit method,as the electrophoresis strips were clear enough.Whereas improved TES dewaxing with water-bath method showed no clear electrophoresis strip.Conclusion Both xylene dewaxing method and DNA extraction kit method are reliable and suitable for DNA extraction from formalin-fixed,paraffin-embedded tissues.更多还原
