ISSR标记鉴别砂梨种质资源的研究
Inter-simple Sequence Repeat Analysis on Pyrus pyrifolia Cultivars Germplasm
【摘要】 为有效区分和利用亲源关系较近的砂梨品种,利用简单重复序列(ISSR)进行多态性、遗传相似性和聚类分析。从100多条ISSR引物对16份繁殖系材料DNA模板进行扩增和电泳检测,结果表明,有16条引物均能扩增出有较高稳定性、重复性和丰富多态性的条带,总数为135条,多态性达70.4%,每条引物扩增3~8条条带;16个砂梨品种间的遗传一致度的变化范围在0.6667~0.9630,遗传距离的变化范围在0.0377~0.4055;‘华高’和‘新高’砂梨品种具有最高的遗传一致度(0.9630),它们之间的遗传距离也最小(0.0377)‘;华高’和‘雪青’砂梨品种的遗传距离最大为0.4044,相应的最低遗传一致度为0.6667。
【Abstract】 In order to efficiently distinguish and use cultivars germplasm of Pyrus pyrifolia with a near genetic relationship,the polymorphism and cluster analysis based on genetic similarity matrices of inter-simple sequence repeat(ISSR) markers were used on 16 cultivars.It showed that 16 pairs of ISSR primes produced 135 amplified fragments of which 70.4%were polymorphic.The average number of alleles per ISSR locus was range from 3 to 8.The genetic similarity coefficients among the 16 cultivars ranged from 0.6667 to 0.9630,and the genetic distance ranged from 0.0377 to 0.4055.The closest genetic similarity coefficients was Pyrus pyrifolia’Niitaka’and’Soumi Tuniitaka’(0.9630) ,the closest genetic distance was 0.0377.Pyrus pyrifolia’ Soumi Tuniitaka’were highly divergent from’Xuqing’,which indicated that the genetic similarity coefficients was 0.667 and the genetic distance was 0.667.
