九州虫草液体培养条件优化
Optimum conditions of liquid culture of Cordyceps kyushuensis
【摘要】 研究九州虫草富锌条件下菌丝体、胞内多糖和胞外多糖含量差异。通过单因子试验、正交试验测定不同因素的作用,菌丝体干重测定采用烘干称重法;胞内和胞外多糖测定应用苯酚-硫酸法和3,5-二硝基水杨酸法。结果表明,九州虫草在加锌条件下菌丝干重最优培养条件是葡萄糖2.0%,蛋白胨1.0%,磷酸二氢钾0.5%,硫酸镁0.5%,硫酸锌200mg/L,装液量80mL/250mL,pH值6.0。最终结果菌丝干重为1.53g/100mL,是原来未加锌基本培养基菌丝干重的2.58倍。
【Abstract】 The differences of mycelia biomass,intracellular polysaccharide and extracellular polysaccharide of Cordyceps kyushuensis were investigated under Zinc-enriched conditions in this paper.The optimal conditions were obtained by single factor and orthogonal tests as followed: sucrose 2.0%,peptone 1.0%,KH2PO4 0.5%,MgSO4 0.5%,ZnSO4 200mg/L,liquid medium volume 80ml/250ml and pH 6.0.Under these conditions,the biomass of mycelia was 1.53g/100ml,which was 2.58 fold higher than that without Zn conditions.
