【摘要】 目的研究真武汤冲剂逆转大鼠慢性压力负荷性心肌肥厚的病理学变化。方法将SD大鼠随机分3组,假手术组8只,手术组和中药组各15只。手术组和中药组行腹主动脉缩窄术,制作心肌肥厚模型。16周行超声心动图,确定术后心肌肥厚,中药组加用真武汤混合饲料喂养,24周手术开胸,心脏组织进行病理学检查,包括HE染色、Masson染色和环氧化酶2免疫组化分析。结果 HE染色中,手术组心肌细胞不同程度增大、核深染,纤维组织增生明显,中药组在不同程度上有所缓解;Masson染色中,手术组胶原纤维增生明显,以血管周围明显,中药组胶原纤维增生较少,与手术组对比,血管周围减少更为明显;免疫组化分析中,假手术组可见散在纤维母细胞,颜色较淡;手术组纤维母细胞不同程度增生,数量增多,体积增大,颜色加深;中药组纤维母细胞轻度增生,数量较假手术组增多,但比手术组明显减少,体积较手术组明显缩小,胞质颜色较假手术组加深。结论真武汤冲剂逆转大鼠压力负荷性心肌肥厚,使心肌细胞增大、核深染、胶原纤维增生的程度减少,且此种改变可能是通过介导环氧化酶2的炎症因子途径实现的。更多还原

【Abstract】 Objective To investigate the pathological changes on reversing myocardial hypertrophy by Zhenwu Tang Granulein rats with chronic pressure-overloaded left ventricular hypertrophy.Methods The experiment was performed in the laboratory of Guangzhou Institute of Respiratory Disease.All the male SD rats were randomized into 3 groups:sham operation group(n=8),operation group(n=15)and herbal medicine group(n=15).The model of myocardial hypertrophy was made by gradually constricting the abdominal aorta.16 weeks later,cardiac ultrasonography was performed in all groups in order to ascertain post-operational left ventricular(LV)hypertrophy.Zhenwu Tang Granule was added in the herbal medicine group with the dose of 12 g/kg in the mixed feedstuff for 8 weeks.In 24th week,we opened the chest by operation and took the cardiac muscle marked by HE and Masson dyeing.The COX-2(cycloxygenase 2)in cardiac muscle was marked by immune histochemistry.Results The myocardial cells enlarged,nucleus deep color and fiber tissue was apparent hyperplasia in different extent by HE in operation group,which was eased in some degree in the herbal medicine group.Collagen fiber was apparent hyperplasia,particularly around the vessel by Masson in operation group but which was reduced in the herbal medicine group.Fibroblast dyed by immunohistochemical was scattered and light colors in sham operation group,which was hyperplastic,growing,volume increasing and dyeing deepen in operation group.Hyperplasia was a little bit in herbal medicine group,which was more than in sham operation group but less than operation group,at the same time there was cutdown in volume compared with operation group and hyperchromatic in cytoplasm to sham operation group.Conclusions Zhenwu Tang Granule could reverse myocardial hypertrophy in rats with pressure-overloaded left ventricular hypertrophy,and ease the degree in which myocardial cell enlargement,nucleus deep color and collagenous fiber.This result maybe happened through the inflammatory factor including COX-2.更多还原
