氧化高密度脂蛋白及其亚型对人脐静脉内皮细胞组织因子途径抑制物-1表达的影响

【摘要】 目的探讨氧化高密度脂蛋白(oxHDL)及其亚型对人脐静脉内皮细胞(HUVECs)组织因子途径抑制物-1(TFPI-1)表达的影响。方法以天然HDL及其亚型作为对照,HUVECs分别经不同浓度(0,10,20,40,80 mg/L)oxHDL、oxHDL2、oxHDL3孵育24 h,逆转录聚合酶链反应(RT-PCR)、Western印迹分别检测TFPI-1 mRNA及蛋白质表达。结果与对照组比较,oxHDL、oxHDL2、oxHDL3能抑制TFPI-1 mRNA和蛋白表达,其中在oxHDL和oxHDL3各系列浓度中以40 mg/L作用最明显,TFPI-1 mRNA和蛋白表达分别下降了28%、35%和49%、47%;在oxHDL2各系列浓度中以80 mg/L作用最明显,TFPI-1 mRNA和蛋白表达均下降了27%,差异均具有统计学意义(P<0.05);且以oxHDL3的作用最强。结论 oxHDL及其亚型能抑制HUVECs TFPI-1 mRNA和蛋白表达,oxHDL3的作用最强。

【Abstract】 Objective To investigate the expression of tissue factor pathway inhibitor-1(TFPI-1) induced by oxidized high densitive lipoprotein(HDL) and its subfractions in human umbilical vein endothelial cells(HUVECs).Methods HUVECs were incubated with oxHDL,oxHDL2,oxHDL3 or their native forms at different concentrations(0,10,20,40,80 mg/L)for 24 h.TFPI-1 mRNA and protein expressions were determined by RT-PCR and Western blot,respectively.Results Compared with control group(ie.HDL,HDL2 or HDL3),oxHDL,oxHDL2,oxHDL3 significantly decreased the expressions of TFPI-1 mRNA and protein,respectively.TFPI-1 mRNA and protein expressions were lowered by oxHDL by 28% and 35%,respectively at the level of 40 mg/L,by oxHDL2 by 27% and 27% at the level of 80 mg/L,and by oxHDL3 by 49% and 47%,respectively,at the level of 40 mg/L(all P<0.05).The effect of oxHDL3 appeared the strongest.Conclusions Ox-HDL and its subfractions could inhibit the expression levels of TFPI-1 mRNA and protein in HUVECs.
