蛇菰水提物对大鼠酒精性肝损伤的保护作用

【摘要】 目的研究蛇菰水提物对大鼠酒精性肝损伤的保护作用,并初步探讨其可能的作用机制。方法健康雄性SD大鼠40只,随机分为对照组、模型组和蛇菰水提物低、高剂量组;模型组和处理组组以酒精灌胃建立大鼠酒精性肝损伤模型,蛇菰水提物低、高剂量组分别给予蛇菰水提物1.5,3.0 g.kg-1.d-1进行预防治疗,10 d后处死全部大鼠,取血测血清谷草转氨酶(AST)和谷丙转氨酶(ALT),肝匀浆测定超氧化物歧化酶(SOD),谷胱苷肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量,肝脏常规切片染色,观察组织学改变。结果蛇菰水提物低、高剂量组组大鼠血清ALT和AST活性明显低于模型组,SOD和GSH-Px活性较模型组升高,MDA含量下降,镜下病理显示蛇菰水提物对大鼠酒精性肝损伤具有保护作用。结论蛇菰水提物对大鼠酒精性肝损伤有显著保护作用。

【Abstract】 Objective To investigate the protective effect of extract from Balanophora spicata Hayata on alcoholic fatty liver disease in rat,and elucidate the possible mechanism.Methods 40 adult SD rats were randomly divided into normal group fed with physiological saline,model group fed with alcohol,low and high dose of extract from Balanophora spicata Hayata groups fed with alcohol and extract from Balanophora spicata Hayata(1.5,3.0 g·kg-1·d-1).After 10 days,the serum alanine aminotransferase(ALT),aspartate aminotransferase(AST) and the contents of malondialdehyde(MDA),glutathione perloxldase(GSH-Px),superoxide dismutase(SOD) in liver homogenate were detected,and the histological changes in liver were evaluated.Results Compared with the model group,extract from Balanophora spicata Hayata significantly decreased the serum transaminase levels and the content of MDA,enhanced SOD and GSH-Px levels in liver homogenate.Histopathological observations were consistent with biochemical changes,suggesting marked protective effect of extract from Balanophora spicata Hayata on acute liver injury in rat.Conclusions Extract from Balanophora spicata Hayata has an obviously protective effect against liver injury induced by alcohol in rat.
