不同年龄女性脂肪组织11βHSD1 mRNA表达水平及临床意义
Expression and clinical significance of 11βHSD1 mRNA in adipose tissues of women in different ages
【摘要】 目的观察儿童期、育龄期及绝经期女性皮下和网膜脂肪组织11βHSD1 mRNA表达水平,探讨人类不同年龄脂肪组织积聚和分布的分子机制。方法取开腹手术女童〔11例,年龄(5.64±3.72)岁〕、育龄期非妊娠妇女〔13例,年龄(33.24±6.25)岁〕及绝经期妇女〔11例,年龄(66.06±7.13)岁〕皮下和网膜脂肪组织。同时测定身高、体重和腰臀围,计算体质指数(BMI)、腰臀比(WHR)。应用实时定量PCR方法测定脂肪组织11βHSD1 mRNA表达水平。结果无论皮下及网膜,女童脂肪组织11βHSD1 mRNA表达水平明显低于育龄期(皮下:P<0.001;网膜:P=0.003)及绝经期妇女(皮下:P<0.001;网膜:P<0.001);育龄期妇女11βHSD1 mRNA表达水平明显低于绝经期妇女(皮下:P<0.001;网膜:P<0.001)。绝经期妇女网膜脂肪组织11βHSD1 mRNA表达明显高于皮下(P=0.01)。成年期女性网膜脂肪组织11βHSD1 mRNA表达水平与BMI(r=0.416,P<0.05),WHR(r=0.775,P<0.001)正相关。结论脂肪组织11βHSD1 mRNA的表达呈现年龄依赖性及部位差异性,并与成年女性BMI、WHR密切相关。提示其可能参与了人类脂肪组织的生长和不同部位分布的调节。
【Abstract】 Objective To observe the expression of 11βHSD1 mRNA in abdominal subcutaneous(Sc) and omental(Om) adipose tissue of the childhood,childbearing ages and postmenopausal women,and the molecular mechanism of the accumulation and distribution of adipose tissue in different ages of human.Methods Sc and Om adipose tissue were obtained from 11 girls aged(5.64±3.72) years,13 women of childbearing ages(aged(33.24±6.25) years) and 11 postmenopausal women aged(66.06±7.13) years.Height,weight,waistline,hipline,BMI and WHR of the objects were determined.The expressions of 11βHSD1 mRNA in different adipose tissues were measured by real time PCR.Results 11βHSD1 mRNA expression in Sc and Om adipose tissue of the girls was significantly lower than that of the childbearing ages(Sc: P<0.00;Om: P=0.003) and postmenopausal women(Sc: P<0.001;Om: P<0.001).11βHSD1 mRNA expression in the women of childbearing ages was significantly lower than that of the postmenopausal women(Sc: P<0.001;Om: P<0.001).In addition,11βHSD1 mRNA expression in Om adipose tissue of postmenopausal women was significantly higher than that of Sc adipose tissue(P=0.01).The expression of 11βHSD1 mRNA in Om adipose tissue of adult females was positively correlated with BMI(r=0.416,P<0.05) and WHR(r=0.775,P<0.001).Conclusions The expression of 11βHSD1 mRNA in adipose tissue shows age-dependent and regional difference,and it is positively correlated with BMI and WHR of adult females,which suggests that it possibly involved in the accumulation and distribution of human adipose tissue.
