异丙酚对急性肺栓塞大鼠炎性因子表达的影响
Effects of propofol on expression of inflammatory factor in acute autoblood pulmonary thromboembolism of rats
【摘要】 目的观察异丙酚对急性肺栓塞(PTE)大鼠炎性因子的影响。方法大鼠分为假手术组、模型组、异丙酚低、中、高剂量(4,8,16 mg/kg)组。制备急性自体血栓PTE模型。进行血气分析;检测肺系数、湿/干重(W/D);放射免疫法检测血液中内皮素(ET)-1、血栓素B2(TXB2)含量;RT-PCR检测肺组织白介素(IL)-1I、L-6、肿瘤坏死因子(TNF)-αmRNA的表达;Western印迹方法检测IL-1I、L-6、TNF-α蛋白的表达。结果异丙酚能显著升高PO2、PCO2水平,降低肺系数、湿/干重、ET-1、TXB2含量,降低IL-1I、L-6、TNF-αmRNA和蛋白表达(P<0.05)。结论异丙酚能抑制炎性因子的表达,对PTE大鼠起保护作用。
【Abstract】 Objective To investigate the effects of propofol on inflammatory factors expressions in acute pulmonary thromboembolism(PTE) of rats.Methods Rats were randomly divided into sham,model and propofol groups(4,8,16 mg/kg).Rat PTE model was established by intravenous injection of auto-blood clots.The PO2 and PCO2 in arterial blood were measured.The coefficient and wet/dry weight(W/D) of lung were detected.The concentrations of ET-1 and TXB2 in plasma were examined by radio-immunity technique.The mRNA expressions of IL-1,IL-6 and TNF-α in lung tissue were assessed by RT-PCR.The protein expressions of IL-1,IL-6 and TNF-α in lung tissue were assessed by Western blot.Results The PO2,PCO2 in arterial blood were significantly increased,while the coefficient,W/D of lung,the contents of ET-1 and TXB2,the expressions of IL-1,IL-6 and TNF-α were significantly decreased after administration with propofol.Conclusions Propofol could inhibit the expressions of inflammation factors and protect PTE rats.
