普洱茶对apoE基因敲除小鼠动脉粥样硬化的影响

【摘要】 目的探讨普洱茶对apoE基因敲除小鼠动脉粥样硬化的影响。方法 40只8周龄apoE基因敲除小鼠,随机分为动脉粥样硬化组、普洱生茶组、普洱熟茶组和血脂康组,每组10只。相同遗传背景的10只同龄同体重同性别的C57BL/6J小鼠为正常对照组。每组均喂以普通饲料,灌胃12 w后,牺牲动物收集静脉血和主动脉。用全自动生化分析仪检测血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C);ELISA检测血清超敏C-反应蛋白(hs-CRP),主动脉经10%福尔马林固定后做形态学观察及图像分析。结果与动脉粥样硬化模型组相比:普洱生茶组与普洱熟茶组血清TG、TC、LDL-C、动脉粥样硬化指数(AI)明显降低;血清hs-CRP含量明显降低;HE染色显示普洱生茶组与普洱熟茶组动脉粥样硬化斑块面积较小和数量较少。结论普洱茶(生茶和熟茶)有明显降低高脂血症和一定抗动脉粥样硬化的作用,且其抗AS作用除与抗脂质过氧化外,可能还与抗炎症有关。

【Abstract】 Objective To observe the effect of puer tea on atherosclerosis(AS) in apoliporotein(apo) E knockout mice.Methods Eight-week-old apoE knockout mice were divided into four groups: AS,puer raw tea,cooked puer and Xuezhikang groups(10 each group).The homogeneous mice of C57BL/ 6J were used as the control.All mice were fed with normal chow diet.After 12 weeks,vessels blood was collected for detecting serum lipid and high sensitive C-reactive protein(hs-CRP).All the mice were filled with 10% formalin and the aortas were stripped for morphological research and photograph analysis.Results Level of serum lipid and hs-CRP in puer tea groups(puer raw tea group and cooked puer group) were lower than those in AS group.Compared with the model group,the AS index of puer raw tea group and cooked puer tea group were decreased obviously.Morphological research showed that AS lesions were found in aortas of model,puer raw tea,and cooked puer group,except normal group.While AS lesions of model group were more serious than those of puer raw tea group and cooked puer group.Conclusions Puer tea can relieve the AS lesions of apoE-gene knockout mice,and it can also inhibit AS development.The mechanism of its anti-inflammation activity is related to modulation of inflammatory factors,which may be a potential way in controlling AS.
