湖羊促性腺激素释放激素受体基因(GnRHR)在消化系统中的表达谱分析

【摘要】 建立湖羊促性腺激素释放激素受体(gonadotropin releasing hormone receptor,GnRHR)基因的实时荧光定量聚合酶链反应(RT-PCR)检测方法,并采用该方法对湖羊消化系统中胃肠各组织的GnRHR mRNA表达水平进行检测。结果表明:GnRHR mRNA在瘤胃、网胃、瓣胃、皱胃、十二指肠、空肠、回肠、盲肠、结肠和直肠部位均有表达,其中瘤胃GnRHR mRNA表达量最高,极显著高于其他各组织(P<0.01);网胃GnRHR mRNA表达量极显著高于十二指肠(P<0.01),显著高于瓣胃、盲肠和直肠(P<0.05);皱胃GnRHR mRNA表达量极显著高于瓣胃、空肠、回肠和盲肠(P<0.01),显著高于结肠和直肠(P<0.05);结肠GnRHR mRNA表达量显著高于十二指肠和直肠(P<0.05)。研究证实了GnRHR mRNA在湖羊消化系统中有表达,这为GnRH功能的多样性研究提供了新的依据。

【Abstract】 To detect the gonadotropin releasing hormone receptor(GnRHR) mRNA expression pattern in sheep’s digestive system,the SYBR green real-time fluorescence quantitative polymerase chain reaction(RT-PCR)method was established.With this method,the GnRHR mRNA expression in stomach and intestinum system of Hu sheep was detected.The results showed that GnRHR mRNA expressions were detected in rumen,reticulum,omasum,abomasums,duodenum,jejunum,ileum,caecum,colon and rectum.The GnRHR mRNA expression in rumen was significantly higher than those in other tissues(P<0.01);the expression in reticulum was significantly higher than that in duodenum(P<0.01) and higher than those in omasum,caecum and rectum(P<0.05);the expression in abomasums was significantly higher than those in omasum,jejunum,ileum and caecum(P<0.01),and higher than those in colon,rectum(P<0.05);the expression in colon was higher than those in duodenum and rectum(P<0.05).It is demonstrated that there are GnRHR mRNA expressions in the digestive system of Hu sheep.It provides new evidences for the research field of GnRH functional diversity.
