电针对大鼠内脏痛结肠壁、背根节和脊髓内乙酰胆碱的影响
Effect of Electroacupuncture on the Ach in the Colon,Dorsal Root Ganglia and Sacral Spinal Cord in the Pelvic Visceral Pain Models of Rats
【摘要】 目的:观察电针治疗内脏痛时结肠壁、背根节和脊髓内胆碱酯酶(AchE)阳性神经元成分的变化,以探讨胆碱酯酶(AchE)阳性神经元成分在痛觉传递中的作用及电针镇痛的机制。方法:26只SD大鼠随机分为正常对照组(6只)、内脏痛组(10只)和电针加内脏痛组(10只)。电针取双侧"足三里"穴,运用酶组织化学技术方法,观察电针对福尔马林诱导的大鼠内脏疼痛时脊髓背根节和结肠壁内胆碱酯酶(AchE)阳性神经元成分的影响。结果:内脏痛组大鼠在注射福尔马林后,结肠壁、背根节和脊髓内胆碱酯酶(AchE)阳性神经元成分显著高于正常组(P<0.01);而电针加内脏痛组,结肠壁、背根节和脊髓内胆碱酯酶(AchE)阳性神经元成分显著低于内脏痛组(P<0.01)。结论:电针对大鼠急性盆腔内脏痛具有预防性治疗作用,Ach可能参与了电针对内脏痛的调节作用。
【Abstract】 Objective:To study the effect of elelctroacupuncture(EA) on the AchE positive neuronal cell bodies and fibers in the colon,dorsal root ganglia and spinal cord in rats with pelvic visceral pain for exploring the role of Ach in pain transmission,and electroacupuncture analgesia.Methods: Twenty-six SD rats were randomly divided into control group(n=6),visceral pain model group(VP group)(n=10) and visceral pain model plus electroacupuncture group(EA+VP group)(n=10).Electroacupuncture(4-6 Hz,dense-sparse waves,0.5-3 V) was applied to bilateral "Zusanli"(ST36) for 60 min.Pelvic visceral pain model was established by local injection of 5% formalin(40μL) into the colonic wall from the anus.Behavioral reactions were observed.After transcardiac perfusion of saline and paraformaldehyde,the rat’s colon near the anus,sacral 2-4 dorsal root ganglia and the corresponding spinal cord segments were removed to be cut into sections(25 μm).The AchE positive neuronal cell bodies and fibers in the colon,dorsal root ganglia and spinal cord were determined by using enzyme histochemistry.Results: In comparison with the contol group,the integral OD values of AchE of the colonic wall,DRG and spinal cord in the model group were significantly higher than those in the control group(P<0.01).Comparison between the model and EA plus VP groups showed that the integral OD values of AchE of the 3 types of tissues in the EA plus VP group were markedly lower than those in the model group(P<0.01),which indicated that EA could suppress pelvic viceral pain(with lower Ach).Conclusion: EA can relieve formalin induced pelvic visceral pain increase the lower Ach produced by visceral pain,which suggests that Ach may be involved in the process of EA analgesia.
