仙客兰组织培养快繁技术的研究

【摘要】 目的:建立仙客来组织培养快速繁殖体系。方法:以仙客来叶片为外植体,消毒后将其置于富含不同激素配比的培养基中培养,筛选各阶段合适的培养基。结果:在MS+6-BA 3.0 mg/L+NAA 0.5 mg/L的培养基组合中,诱导愈伤率达到80%,增殖培养以MS+6-BA3.5 mg/L+NAA0.5 mg/L为最好,可达到10倍以上。MS+NAA3.0 mg/L培养基为最佳诱导生根培养基,生根苗移栽成活达到90%。结论:实验初步建立了仙客来的组培快繁体系。

【Abstract】 Objective: To developed a rapid micropropagation system for Cyclamen persicum Mill.Methods: The rhizomes that had sprouted of Cyclamen persicum as explants were cultured on MS medium supplemented with different concentrations of BA in combination with different concentrations of NAA or 2,4-D to select optimized medium for different phase.Results: The optimum induction medium was MS + 6-BA 3.0 mg/L + NAA 0.5 mg/L and the callus induction rate can reach 80 percent,The solid MS medium was MS + 6-BA 3.5 mg/L + NAA 0.5 mg/L was suitable for the multiplication of shoots and the proliferation can reach more than 10 times.The optimum rooting medium was MS + NAA 3.0 mg/L was suitable for the shoots rooting and 100% plantlets with roots were obtained.Conclusions: The results can used to further study of Cyclamen persicum Mill rapid propagation.
