玻璃化冷冻羊皮异种移植后血清白细胞介素-4和干扰素-γ表达的研究

【摘要】 目的了解玻璃化冷冻羊皮异种移植到大鼠创面后,大鼠血清中白细胞介素4(IL-4)和干扰素-γ(IFN-γ)的表达特点。方法 80只大鼠背部设计制作4cm×4cm大小全层皮肤缺损创面,将玻璃化冷冻羊皮与大鼠自体微粒皮混合移植于大鼠创面上作为实验组,40只大鼠异体皮与自体微粒皮混合移植作为对照组,对比观察两组创面愈合情况,分别于术后1、2、3、4周末收集大鼠血清,检测IL-4和IFN-γ的值进行对比。结果 4周后对照组的微粒皮均扩展覆盖创面,而实验组大部分创面愈合,微粒皮虽有扩展但没有完全覆盖创面。大鼠血清中IL-4和IFN-γ的表达方面,实验组和对照组施行移植术后血清IFN-γ和IL-4均随时间逐渐增高,术后至2周末,两组血清IFN-γ和IL-4表达差异均无统计学意义(P>0.05),术后3周末,实验组血清IL-4和IFN-γ表达明显高于对照组,两组间差异有统计学意义(P<0.05)。术后4周末,对照组创面基本愈合,实验组大部分创面已愈合,两组的羊皮均被完全排斥,实验组和对照组大鼠血清IL-4和IFN-γ的表达均下降,两组相比,差异无统计学意义(P>0.05)。对比IFN-γ/IL-4的值在四周内两组间无统计学意义(P>0.05)。结论羊皮移植于大鼠体表创面后两周的时间内,冷冻羊皮与异体皮有相似的免疫排斥反应特点,其对微粒皮的保护与异体皮有相似的效果。

【Abstract】 Objective To learn the expressional feature of IL-4 and IFN-γ in rats serum after vitrification cryopreservation sheep skin was transplanted on wound of rats. Methods In the experimental group, vitrification cryopreservation sheep skin and microskin of rats were confused transplant on wound of rats.while in the control group, allosome skin and microskin of rats were confused transplant on wound of rats.The wound healing time was compared and rats seruam at 1 st、2 rd、3 rd、4 rd weekend were collected, to check and compare the expression levels of IL-4 and IFN-γ between two groups. Results The microskin of two groups all was apparently expanded, but the wound healing rate of experimental group was lower than the control group. The wounds of control group were all cured, but a few of wounds of experimental group were not cured. In both experimental group and control group, the expression levels of IL-4 and IFN-γ increased with time passed by. At the second weekend after transplantation, the expression levels of IL-4 and IFN-γ had no significant differences between two groups. At the third weekend, the expression level of IL-4 of experimental group was obviousely increased, with significant difference between two groups. At the fourth weekend, because the sheep skin has been repelled by body of rats, the expression levels of IL-4 and IFN-γ became lower and had no significant differences between two groups. There was no significant difference about the ratio of IFN-γ and IL-4 between two groups. Conclusions During two weeks after transplant ation, vitrification cryopreservation sheep skin and allosome skin had same immune rejection reaction feature.
