紫杉醇聚氰基丙烯酸正丁酯纳米粒对人卵巢癌细胞的毒性
Study on the cytotoxicity of paclitaxel-loaded polybutylcyanoacrylate nanoparticles on human ovary cancer
【摘要】 目的:评价制备紫杉醇聚氰基丙烯酸正丁酯纳米粒(PTX-PBCA-NPs)的原料的生物安全性以及PTX-PBCA-NPs的细胞毒性。方法:采用四噻唑蓝法(MTT法)和检测乳酸脱氢酶(LDH)活性的方法考察空白PBCA-NPs及其聚合的原料、PTX-PBCA-NPs对L-02人正常肝细胞、卵巢癌敏感株(A2780)和卵巢癌耐紫杉醇肿瘤细胞株(A2780/T)的细胞毒性。结果:制备的空白PBCA-NPs只有在大于608 ng·mL-1时,对于L-02细胞具有明显的毒性(P<0.05);在质量浓度304~608 ng·mL-1,空白PBCA-NPs对A2780和A2780/T细胞有明显毒性(P<0.05)。与同一浓度PTX溶液比较,PTX-PBCA-NPs对A2780和A2780/T细胞的毒性作用明显(P<0.05)。结论:空白PBCA-NPs有一定的生物安全性,PTX-PBCA-NPs在对卵巢癌肿瘤细胞有一定的杀伤能力。
【Abstract】 OBJECTIVE To investigate the toxicity of paclitaxel-loaded polybutylcyanoacrylate nanoparticles(PTX-PBCA-NPs) and its materials on the L-02 cell,A2780 cell and A2780/T cell.METHODS The cytotoxicity of blank PBCA-NPs,its materials and PTX-PBCA-NPs was measured by MTT and LDH assay.RESULTS The toxicity of blank PBCA-NPs on A2780 cell and A2780/T cell was greater than L-02 with a concentration range of 304-608 ng·mL-1.In addition,the drug formulated in PTX-PBCA-NPs showed a greater cytotoxicity than paclitaxel against A2780 and A2780/T cells.CONCLUSION It was concluded that the blank PBCA-NPs had better biological security,less cytotoxicity;and PTX-PBCA-NPs had good anti-tumor activity.So PTX-PBCA-NPs would be a novel paclitaxel preparation in clinic for the treatment of tumor.
